Effect Of Serum Containing Core Of Jianpi Xiaoai Recipe On Angiogenesis Of Chicken Chorioallantoic Membrane

Objective:By studying the influence of serum containing the core of Jianpi Xiaoai Recipe on angiogenesis of chick embryo chorioallantoic to clarify the recipe's mechanism to reduce the rate of recurrence or metastasis of colorectal cancer.Methods:1.Eggs incubated for 7d were made into CAM models,then divided them into five groups randomly, named as high dose of Jianpi Xiaoai Recipe group, medium dose of Jianpi Xiaoai Recipe group, low dose of Jianpi Xiaoai Recipe group, blank serum group,the positive drug control group of Shenyi capsule and the positive drug control group of Endostar, 15 eggs in each group. 2.Added high,medium,low dose serum containing Jianpi Xiaoai Recipe,blank serum, serum containing Shenyi capsule and Endostar injection respectively,and incubated eggs for 3d continuely.3.Fixed the chorioallantoic membrane with Paraformaldehyde solution, then observed and cut off the specimens with the scissor used in Ophthamology,tiled the specimens on slides.4. After the water on the specimens evaporating,took photos of the specimens from the same distance with digital camera,and acquisited images under the optical microscope.5.Using Image-Pro Plus 6.0 image analysis software,quantitatively calculated and compared the ratio of blood vessel area of each group.Results:The ratios of blood vessel areas of Jianpi Xiaoai Recipe groups were less than that of blank serum group(P<0.05),and the ratios of blood vessel areas of high,medium dose groups were less than that of low dose group(P<0.05), but there was no significant difference between hign dose group and medium dose group(P>0.05), the ratios of blood vessel areas of high,medium dose groups were more than that of Endostar group(P<0.05),while the differences between the high dose of Jianpi Xiaoai Recipe group, medium dose of Jianpi Xiaoai Recipe group and the positive drug control group of Shenyi capsule were of no statistical significance(P>0.05).Conclusion: Serum containing core of Jianpi Xiaoai Recipe can inhibit angiogenesis of CAM to a certain extent, though the inhibition effect of Serum containing core of Jianpi Xiaoai Recipe on angiogenesis of CAM is weaker than that of Endostar,while under the condition of medium and high dose,its inhibition effect on angiogenesis is equa to that of Serum containing Shenyi capsule.It indicates that Jianpi Xiaoai Recipe may inhibit tumor angiogenesis to achieve anti colorectal cancer recurrence and metastasis effect.
