| ObjectiveTo explore the effects of Qingre Jiedu Huazhuo tablets on tissue morphology of liver and small intestine,liver function(ALT, AST, ALP, GGT, TBIL, DBIL, IBIL,propagated, GLOB), endotoxin level, TNF- α, α-SMA,ZO-1,Occludin on model rats of liver injury on IETM,which induced by alcohol and high fat diets.Methods50 rats were adaptability to feed the first 5 days,then grouped according to random number table method.10 rats were used as blank control group,which fed with ordinary diet and distilled water(Dose of 1 ml / 100 g,Weighed twice weekly to adjust the dosage),others fed with 52 degrees of liquor and high fat diet as model group.All the rats were cut the tail blood to test the endotoxin level and liver function at the end of 3th month,when the model were established,the successful model rats were randomly divided into 3 groups(model group,experimental group 1,experimental group 2), model group were treated with liquor and distilled water,experimental group1 were treated with liquor and Qingre Jiedu Huazhuo tablets,experimental group 2treated with distilled water and Qingre Jiedu Huazhuo tablets,blank control group treated with distilled water.twice a day(9AM/4PM) for 1 month.Results1.The liver pathological of model group rats obvious fatty degeneration and inflammatory change,even a few change of liver fibrosis,the fatty degeneration of experimental group 1&2 were significantly reduced,no significant inflammation and liver fibrosis either.The small intestine pathologica of model control group significantly inflammatory change, after treatment, experimental group 1&2inflammatory changes were significantly reduced.2.The endotoxin level of model group were obviously higher than blank control group and experimental group 2(P < 0.01),experimental group 2 was lower thangroup 1(P<0.05).3.Both experimental group 1&2’ liver function(ALT、AST、GGT) were lower than model group(P < 0.01),experimental group 2’ALT and AST were lower than group 1(P < 0.01),experimental group 2’ALP was lower than group 1(P <0.05),experimental group 2’DBIL was lower than group 1 and model group(P<0.05).4.The TNF-α level of experimental group 1&2 were lower than model group(P<0.05).The α-SMA level of experimental group 2 was lower than model group(P<0.05).5.The Occludin level of experimental group 2(P <0.01)was obviously higher than model group,so did group 1(P<0.05). Both experimental group 1&2’ Occludin level were higher than model group(P<0.05).6.There was a negative correlation between TNF-α and Occludin(r=-0.543,P<0.01),so was ALT and Occludin(r=-0.595,P<0.01);There was a positive correlation between TNF-α and endotoxin levels(r=0.603,P<0.01),so was TNF-α and α-SMA(r=0.592,P<0.01).Conclusion1.In terms of morphology,Qingre Jiedu Huazhuo tablets can obviously relieve the fatty degeneration and inflammatory changes of liver tissue, also can relieve the inflammatory changes of intestinal tissue.2.Qingre Jiedu Huazhuo tablets have effects on improving liver function,reducing endotoxin levels, inhibiting inflammation and liver fibrosis.3.The mechanism may be related to increasing Occludin and ZO-1 levels of intestinal tissue,protecting intestinal barrier function,reduce the absorption of endotoxin,terminating the vicious cycle between intestinal endotoxemia and liver injury. |
