Objective:From the blood cell analysis, pathology, immuno-histchemistry, and other research Qingrejiedu Huazhuo film endotoxic liver injury in rabbits of TLR4, NF-κB signaling pathway downstream effectors of TLR4/NF-κB IL-2, IL-6IL-8,1L-10as well as blood count, liver function, serum endotoxin level of impact, investigate the mechanism of action of of Qingrejiedu Huazhuo film.Methods:In this study,25healthy New Zealand rabbits were completely randomized into five groups, namely the blank group, model group of Phase1, Phase1Qingrejiedu Huazhuo piece group (hereinafter referred to as a the stage Huazhuo chipset),2-stage model group,2the the stage Qingrejiedu Huazhuo chipset (hereinafter referred to as Phase2piece Huazhuo group)(n=5). Phase1model group, model group of Phase2and Phase1Huazhuo piece group, two-stage Huazhuo piece group along the ear vein injection of E. coli toxin10μg/ml/kg, blank control group was injected1ml/kg saline. Blood, heart blood test liver function, CRP and IL-2, IL-6, IL-8,1L-10, take liver tissue HE staining of liver tissue morphology pathological changes observed by immunohistochemical method in each group liver of TLR4, NF-κB expression.Results:(1),2-stage Huazhuo piece group with two stage model group, the WBC rise, has a significant (p<0.05).1phase Huazhuo piece group with a stage model group, the WBC rose has a significant (p<0.05).1phase Huazhuo piece group with a stage model group than in the Phase2Huazhuo piece group with two stage model group, the endotoxin levels were decreased, there was significant (P<0.05);(2)Huazhuo piece for the stage2group with two stage model group than IL-2concentrations were significantly decreased gap significantly significance (p<0.05);2stage Huazhuo piece group with two-stage model group than IL-2concentrations were significantly decline, the difference was significant (p<0.05);2stage Huazhuo piece group with two-stage model group than IL-6concentrations were significantly decreased, the difference was significant (p <0.05);2the stage Huazhuo piece group of IL-8concentration was significantly decreased compared with the phase2model group difference has a significant (p-<0.05);2stage Huazhuo piece group with stage2model group than that, of IL-10concentration was significantly decreased, the difference was significant (p<0.05);2stage Huazhuo piece group with two-stage model group than CRP decreased significantly, the difference was significant (p<0.05); stage1Huazhuo piece group with a stage model of group, CRP was no significant decline;(3), The1phase Huazhuo piece group and two stage Huazhuo chipset TLR4, NF-κBp expression significantly reduced compared with the model group, a stage Huazhuo piece group with a stage model of group of TLR4average optical density decreased, the gap has significantlysignificance (p<0.05); Huazhuo piece for the stage2group with two-stage model of group of NF-κBp65in the average optical density decreased, the gap has a significant (p<0.01). The Phase2Huazhuo piece group compared with stage1Huazhuo chipset in the expression of NF-KBp65decreased more significantly.Conclusion:1. Qingrejiedu Huazhuo-chip ETM liver injury may prevent the inflammatory response and clear to endotoxin;2o The of Qingrejiedu Huazhuo piece ETM liver injury may be down TLR4, NF-κappa B expression;3.Qingrejiedu Huazhuo tablets may regulate CRP and TLR4/NF-κB signal transduction pathway downstream of IL-2, IL-6, IL-8and IL-10is related to the role of ETM liver injury.
