Purpose: To investigate the effect of sijunzitang(sjzt) decoction on the fecal bacteria,intestinal mucosal morphology, macrophage phagocytic function and IFN-? expression of digestive tract infection in mice with spleen deficiency syndrome.For the prevention and treatment of spleen deficiency syndrome of Candida infection to provides a theoretical basis.Material and method: This experiment SPF kunming mice were randoml y divided into five groups, that is normal control group(A)? spleen model group(B)?spleen+Candida albicans group(C)?spleen+Candida albicans +flucona zole group(D)?spleen+Candida albicans+sjzt group(E).The mice were fed for one week after modeling.In addition to the normal control group(A), the other four groups of mice with spleen deficiency take complex factors, continuous modeling 14 d.The 15 d,mice of group C?D?E gavage for Candida albicans(2×108CFU/m L)suspension 0.5m L/only,While giving group A?B Physiological saline 0.5 m L/only.The 16 d, group D were fed with fluconazole,group E micew erefed with sjzt.Once a day,for a week.Observe the general state of the mice in each group.In start lavage sjzt decoction 7, 14, 21 days, detectionof Candida albicans in feces.The jejunum of mice were prepared with HE staining and path ological observation of intestinal mucosal morphological changes.Determination the content of mice serum IFN-?with ELISA method.The expression level in small intestine of mice was detected by Western blotting.The level of IFN-? m RNA examined the expression of small intestineof mice by RT-PCR.Results: 1. Effect of Sjzt decoction on the general state of intestinal infection of Candida albicans in mice with spleen deficiencyCompared with the normal control group,spleen model group?spleen+ Candida albicans group the experiment of 22 day, food less poor appetite, hair withered and yellow, body weight and food intake decreased significantly, dull eyes is very narrow.The experiment of 29 day,spleen+Candida albicans group mice weight decreased significantly,the ear color pale ear after hair scattereddull,will dilute and prolapse of the anus,weak limbs swaying walk, arch back,afraid of the cold,activities reduce hi curled up together.Compared with the spleen+ Candida albicans group,spleen+Candida albicans +fluconazole group?spleen+Can dida albicans+sjzt group after administration,the general state of mice has impro ved,the 22 day of the experiment began to rise gradually,the 29 day of the Experiment,the basic shape of feces,food intake increased and so on.2. Sjzt decoction can improve the general state of mice with spleen deficiency infection of Candida albicans,after sjzt decoction, the amount of fecal bacteria was also significantly decreased. 2. Sjzt decoction can reduce the inflammation of the intestinal mucosa of spleen deficiency mice infected with Candida albicans. 3. Sjzt decoction can improve the spleen deficiency of mice infected with Cand ida albicans of peritoneal macrophage phagocytosis ability of Candida albicans. 4. Sjzt decoction could increased the spleen deficiency mice infected with Candida albicans,the expression level of IFN-?protein and m RNA in serum of IFN-?and IFN-?in small intestine.
