| 【objective】: by studying with a kind of traditional Chinese medicine for activating blood circulation(red paeony root,angelica,turmeric na)on knee osteoarthritis rabbit insulin factors,transforming growth factor beta 1,discuss promoting blood circulation to remove blood stasis type of the mechanism of action of traditional Chinese medicine in treating knee osteoarthritis.【Method】: take 48 healthy New Zealand rabbit ears,only 36 rabbit Hulth build mode method is adopted to establish the rabbit model of knee osteoarthritis,after the success of the building will be randomly divided into control group,the Chinese medicine group(high concentration and low concentration of traditional Chinese medicine two variables),model group,12 each.The remaining 12 for control.After the success of the building in order to do the corresponding processing,put to death the experimental rabbit after 4 weeks,after taking joint effusion after processing the application ELASA kit method respectively to detect the contents of IGF1 and TGF-beta,and rabbit chondrocytes after specimen observation and MARKIN score.)Statistical processing: using SPSS13.0 for WINDOWS for statistical analysis.The numerical results by x + s to P < 0.05 for the difference was statistically significant,P < 0.01 there are significant differences.【Results】:(1)the cartilage cells on the cartilage general observation:(1)the control of the rabbit knee joint has no obvious joint effusion,cartilage surface smooth appearance is blue and white,deep and bright color,basic no crack,structure of cartilage Mankin ’s score of 0 points.Model group basically a joint effusion,left knee joint articular surface is rough,even some parts appear erosion,all of the cartilage surface yellowish gray,significantly lower gloss,slightly rough surface,the more coarse;Traditional Chinese medicine group is only part of the rabbitknee joint effusion and synovial hyperplasia,cartilage change still is given priority to with rough,but to a lesser degree than model group.Chinese medicine group and model group by Mankin cartilage structure ’s score comparison,the difference was statistically significant(P < 0.05).(2)after hematoxylin-eosin staining observation: basic control staining showed cartilage surface level off,the size of the cartilage layer symmetry;Cell size is consistent,the tide line good integrity and toluidine blue stain evenly distributed according to law;With visible cartilage after dyeing model group was obviously thinning,cartilage surface cracks,cartilage cells decreased significantly;Traditional Chinese medicine(TCM)observed group distribution of cartilage cells is neat and cell count increased in the model group,cartilage surface relatively flat,fissure,less cartilage thickness thickness in the model group,there are tide line.Control group and model group,positive drug group by Mankin ’s score comparison with the traditional Chinese medicine group,the difference had statistical significance(P < 0.05).(2)on cytokines: insulin growth factor 1: the content of each factor comparison,control group and model group,the Chinese medicine group and control group differences by comparing all have statistical significance(P < 0.05),model group compared with the traditional Chinese medicine group and the control group difference was statistically significant(P < 0.05),Chinese medicine group compared with control group there was no statistically significant difference(P > 0.05)【conclusion】: the kind of Chinese traditional medicine for activating blood circulation can inhibit bone hyperplasia,its action can be through regulating the insulin-like growth factor 1. |
