| BackgroundDiabetes mellitus(DM)is a group of metabolic diseases caused by many factors such as genetic factors,environmental factors and so on,which is characterized by chronic hyperglycemia.It is caused by the defect of insulin secretion and/or function.Its etiology and pathogenesis have not been clearly explained.Its prevalence is high,and it is increasing year by year along with the improvement of living standard.Now it has become a chronic non communicable disease which seriously endangers people’s health after cardiovascular,cerebrovascular disease and tumor.Among diabetics,type 2 diabetes(T2DM)is the most common,accounting for about 90%-95%of the total number of patients.At present,studies have shown that the pathophysiology of T2DM is relative lack of insulin secretion caused by insulin resistance(IR)and pancreatic beta cell dysfunction.IR is the common pathological basis of many clinical chronic diseases,such as diabetes,hypertension,lipid metabolism disorder and so on.In recent years studies have shown that inflammation and the occurrence and development of IR is closely related to inflammatory cytokines such as tumor necrosis factor alpha(tumor necrosis factor alpha,TNF-alpha),interleukin 6(IL-6),high sensitive CRP(hs-CRP)in IR and T2DM in the occurrence and development of a link for.Among them,the inflammatory factor TNF-alpha is confirmed to be crucial in IR.It has been proved that traditional Chinese medicine has many advantages in improving IR,treating T2DM and its complications with multiple targets,multiple paths,and safety without obvious toxic and side effects.It has been confirmed by regulating the level of inflammatory factors.The clinical practice shows that Huang jing has good effect on reducing sugar and fat,but its influence on TNF-alpha and its correlation with the theory of inflammation are still blank.ObjectiveIn this study,we studied the effect of Rhizoma Polygonatum on the level of TNF-alpha and its effects on IR and glucose and lipid metabolism disorders in type 2 diabetic rats,and explored the correlation between them,aiming to apply the theory of inflammation to clinical practice.Methods(1)Forty SD rats were selected as the subjects.The rats were fed with high sugar and high-fat diet combined with low dose streptozotocin.The T2DM rat model was established,and rats were divided into groups,includingblank control group,model group,Chinese medicine group,western medicine group.(2)This study was aimed to determine the total cholesterol of rats(TC),triglyceride(TG),low density lipoprotein(LDL-C),high density lipoprotein(HDL-C),fasting blood glucose(FPG),fasting insulin(FINS),atherosclerosis index(IAI)and calculate the insulin resistance index(HOMA-IRI)and observe the pathological sections of rats.With the help of Excel software to establish a database,data aggregation in rats,SPSS20.0 software was used for statistical methods,nonparametric test,chi square test to compare the difference.P<0.05,indicates the difference was statistically significant,P<0.01 indicates that there are significant differences.Result(1)Huang Jing can significantly regulates the blood glucose metabolism in T2DM rats,and reduce the level of fasting blood glucose in rats.(2)Huang Jing can significantly improved lipid metabolism in T2DM rats,which could effectively reduce total cholesterol,triglyceride,LDL and atherogenic index in rats,indicating that it has some preventive and therapeutic effects on cardiovascular and cerebrovascular diseases and diabetic complications.(3)Huang Jing can reduce the IR state by repairing the damaged insulin signaling pathway by reducing the secretion of TNF-alpha in adipocyte and improving the metabolic disorder of glycolipid.(4)Huang Jing has no obvious effect on HDL in T2DM rats. |
