Nucleo-cytoplamic transport plays a very important role in eukaryotic cells for their life activities.It involves a complex interaction between karyopherins and nuclear pore complex(NPC)and its cargos,and need the RanGTP concentration gradient.There are two importins,Kapl and Kapp2,that have been well studied.They are able to recognize their own type of nuclear localization signals:Kap?1 for classical NLS and Kap?2 for PY-NLS respectly.In plants,however,the research about Kap?2 is few.This research verifies the interaction between certain proteins and AtTRNl(AtKap?2)by yeast two-hybrid on the basis of the Arabidopsis cDNA library that has been established and the results acquired by screening the library in our laboratory;searches the sequences that may be recognized by AtTRNl by means of the bioinformatics methods and further verifying the interaction between the truncated proteins and AtTRNl.Results:First we obtain 18 proteins that can interact with AtTRNl,which consist of 2 Ran proteins,many enzymes and transcription factors,some regulatory proteins,transporters and 1 protein uncharacterized.Second,the results of truncated experiment suggest AtTRNl may be inclined to recognize bipartite NLS,however,that how AtTRNl does recognize proteins without bipartite NLS remains unknown.Third,the NLS of AtTRNl' cargos have no overall basic characteristics.high structure disorder and low sequence similarity.Through the research of this paper,we obtain several differences between AtTRNl and mammalian KapP2 in their cargos and binding sequences,and provide a good basis for studying function and regulatory mechanism of AtTRNl.
