In recent years,looking for secondary metabolic products with high efficiency and low toxicityfrom marine sponges is one of the hot research spots in marine drugs.Marine sponge Agelas are sessile animals of Phylum Porigera, Subclass Demospongiae, Class Agelasida, family Agelasidae. The genus contains 41 species, andnatural products from 19 species have been systematically studied by domestic and foreign scholars. The chemical constituents mainly include pyrrole alkaloids and diterpene alkaloids with good bioactivities, such as antimircrobial, antimalarial, antileukemic, cytotoxic, antifouling activities, as well as inhibitory and activation effects on some enzymics. The South China Sea locates in the South of Tropic of Cancer and belongs to the tropical monsoon climate, Therefore, there are a large number of marine biological resources with strong relatively metabolisms. In order to search for natural products with pronounced biological activities and unique structural features, we have performed an investigation on the chemical constituents and biological activities ofthe dichloromethane/methanol fractionof the sponge Agelas aff. Nemoechinata collected from the Paracel Islands of the south of China, in November, 2009.Isolation and structure determination of secondry metabolites:Using TLC, normal phase chromatography, reversed phase chromate-graphy, Sephadex LH-20, MPLC and HPLC, 17 compounds were isolated from dichloromethane/methanol partof the sponge Agelas aff. Nemoechinataand 13 compounds were identified as Nemoechine F(1**, new compound), Nemoechine G(2**, new compound), Debromomukanadin B(3*, new natural compound), Nemoechine C(4), Nemoechine E(5), 8'-oxoagelasine D(6), agelasine B(7), 2-oxo-agelasine B(8), 2'-oxo-agelasidine C(9), 1H-Pyrrole-2-carboxylic acid ion(1-)(10), 2-pyrrole-carboxamide(11), 9-Methyladenine(12) and Uracil(13)on the basis of spectral methods such as UV, IR, NMR and MS.Biological activitiesof isolated compounds: The cytotoxicities against HCT-116(Human colon cancer cells), He La(human cervical carcinoma cells), Jurkat(Human T lymphoid leukemia cells), Hep G2(Human hepatocellular carcinoma cells), MCF-7(human breast cancer cell), A-549(Human lung cancer cells) and K562(chronic myelogenous leukemia cells)cancer cell lines of the isolated compounds were evaluated by SRB or MTTmethods. In addition, their antimircrobial activities against Staphyloccocus aureus and Bibrio Parahemolyticus weredetected using antimicrobial susceptibility test. The results indicated that all compound showed week activity against Staphyloccocus aureus and Bibrio Parahemolyticus. Besides that, compound 2 showed week activity against Jurkat cell lines with IC50 value of 17.1?M and compound 6 showed week activity against Hela cell lines with IC50 value of 11.438 ?M.Research significance:The investigation on the chemical constituents and biological activities ofdichloromethane/methanol partof the sponge Agelas aff. Nemoechinataenriches the database of sponge secondary metabolites, and it also provided important evidences for development of marine resources.
