In China,lotus seeds have a long history of cultivation,which is mainly cultivated with underground lotus root.We are familiar with the Chinese lotus seed varieties,like Jianlian,Ganlian,Xianglian and Xuanlian[1,2].Lotus seeds are rich in protein and other trace elements.Most of the cultivated areas of lotus are in East China,South China and central china waters.And the lotus seed industry is still a lack of scientific processing industrial chain,its commercial value not excavated better.So the purpose of this study is designed to white lotus as the raw material,produce a rich nutrition and unique flavor of lotus seed protein drinks,and achieve a certain output of factory production.1)Optimization of enzymatic hydrolysis conditions of two enzymes.Choose two kinds of medium temperature?-amylase,under the condition of the determined paste liquid ratio,The other four single factor like enzyme solution temperature,adding quantity,enzyme digestion time and solution PH were tested.According to the results of single factor experiment,we designed the orthogonal test.the results showed that a-amylase A has the optimal conditions for lotus seed starch enzyme solution:5%of enzyme addition,30 min of enzymolysis time,60?of enzymolysis temperature,PH6.2 of enzymolysis;optimum conditions of?-amylase B for lotus seed starch enzyme solution:0.5%of enzyme addition,45min of enzymolysis time,40?of enzymolysis temperature,pH 5.5 of enzymolysis.Two enzymes were tested,and the optimum enzymolysis conditions found the degree of enzymatic hydrolysisof a-amylase B is significantly higher than a-amylase A,reached 25.99±0.62 mg/mL.And compared with the enzyme solution,the residual amount remaining lotus seed residue in enzymatic hydrolysis solution of?-amylase B was less than?-amylase A.The results indicated that the?-amylase B enzymatic hydrolysis more completely,so the?-amylase B was selected to carry out the limited enzymatic hydrolysis experiment.2)Limited enzymatic hydrolysis of lotus starch.We select?-amylase B to the following test.When control the other optimal enzymatic hydrolysis conditions constant,beverage lotus seed centrifugal sedimentation rate decreased with the increase of the enzymatic hydrolysis time.The precipitation rate of the beverage tends to be constant the enzymatic hydrolysis of 40min.By comparing these product sensory score of beverage under different enzymatic hydrolysis time,we found in the enzymatic hydrolysis of 40 min sensory evaluation score was higher.so 0.5%of enzyme addition,40min of enzymolysis time,50?of enzymolysis temperature,PH5.5 of enzymolysis was considered as the degree enzymatic hydrolysis control point.And the content of reducing sugar under the control point in the enzymatic solution was 23.76±0.38 mg/mL.3)Analysis of protein contentThe?-amylase B was chosen in protein content analysis.By measuring the protein content of the precipitated of lotus seed beverage,the protein content was as high than 69.56±0.14%,which indicated that a large part of the precipitation in the beverage was caused by the protein without adding the stabilizer.At the same time,the protein in lotus seed residue was 52.37±0.19%of the protein in lotus seeds,which indicated that about half of the protein was still in the lotus.So we need to add a certain amount of protease in enzymatic hydrolysis process,and thus enhanced the nutritional value of beverage.4)Determination of compound stabilizer for lotus seed beverage.Using sodium carboxymethyl cellulose and glycerol monostearate in the lotus seed beverage compound stabilizer experiment.In the factor experiment showed that0.09%sodium carboxymethyl cellulose and 0.12%monoglyceride has the less amount of centrifugal precipitation respectively.The compound stabilizer experiment results show that the stability of lotus seed beverage is better when 0.09%sodium carboxymethyl cellulose and the mixture of 0.09%monoglyceride was added.5)Plant design on a yearly production of 20000 tons of lotus seed beverage.According to the results,referring to the plant design requirements and using of optimized process parameters,We made a design on lotus seed beverage modern production process,did a research on the equipment selection,calculated the material balance and take food safety issues into consideration,finally through economic analysis,we drew the factory blueprin,which could be used in lotus seed beverage modern production.
