Flow injection chemiluminescence(Fl-CL)is a simple,sensitive and low-cost method for rapid determination.This method has been widely applied to diverse areas like pesticides,environment,food,pharmaceuticals,etc.Flow injection chemiluminescence immunoassay(FI-CLIA)has been established by combining flow injection chemiluminescence with immunological method based on the specific reaction between the antibody and antigen.It exhibits some significant advantages of high sensitivity and specificity,simple sample preparation,low cost and automatic operation.In this dissertation,flow injection chemiluminescence immunoassays for the detection of small molecule diclofenac and Sudan I were established.The dissertation includes four aspects:In chapter 1,the background and significance of the research have been introduced,including the principle of chemiluminescence and imnunoassay,immunosensor,flow injection chemiluminescence immunoassay,solid phase materials,nanoparticles,etc.In chapter 2,the chemiluminescence signal was effectively amplified by the introduction of carboxyl resin beads with large specific surface area,a highly specific polyclonal antibody and the second antibody labelled with horseradish peroxidase.A flow injection chemiluminescence competitive immunoassay for the detection of diclofenac was established.In chapter 3,a flow injection chemiluminescence immunoassay based on glass slide was proposed.Coating antigen could be immobilized on glass slide by modifying graphene oxide chitosan membrane.It provides a new way to build biosensor,and it can be firther applied to study the detection of Sudam Ⅰ and other small molecules.In chapter 4,silica nanoparticles with good dispersion and homogeneous size were prqpared.The surface of silica can be modified by tiree steps so that horseradish peroxidase can be successfully modified.The chemiluminescence signal can be amplified effectively by using silica nanoparticles modified with horseradish peroxidase and altibody as probes.
