In this work,the anti-allergic activities of six kinds of flavonoids were studied by RBL-2H3 cells in vitro.Ginsen flavonoids SA and SB were observed to be the right ones with strongest inhibitory effects on the releasing of ?-HEX of RBL-2H3 cells.Then the high-speed counter-current chromatography were applied to purify the flavonoids.In the end,the quantitive analyses were investigated in ginseng plants.As follows:1.Anti-allergic activities of flavonoids in vitro.The anti-allergic effects were investigated by RBL-2H3 cell lines,the release of ?-HEX was used to evaluate the effects of flavonoids(rutin,quercetin,murraya flavonoids JA,JB,ginseng flavonoids SA,SB).As the results shown,Ginseng flavonoids SA and SB showed the lowest ?-HEX releasing,nearly 68.83% and 65.40%,at the concentration of 25 ?mol/L.2.Extraction,purification and identification of the ginseng flavonoids SA and SB.The stems and leaves of Panax were selected as raw materials to optimize experimental condition,the best extraction process was obtained.As follows: reflux extraction by methanol,liquid material ratio 10:1,three times,1.5 h,1 h and 0.5 h respectively.The total extract were separated by AB-8 resin,the total flavonoids were obtained.High-speed counter-current chromatography was used to purify flavonoids.Two flavonoids were obtained with purification of 98.2% and 96.7%,they were identified by NMR,according to the literature,they were ginseng flavonoid SA and SB.3.Quantitative analysis of ginsen flavonoids SA and SB in Panax plant.According to the results,the peak area(mAUs)and concentration(mg/mL)showed a good linear relationship,precision,repeatability and stability.The RSD values were less than 2%,indicating the content determination method has high precision and strong repeatability,good stability,and accuracy.The content of ginsenosid SA and SB in Panax plants(Panax,Panax stems and leaves,ginseng,ginseng leaves and American ginseng)were determned by HPLC.The results showed that the content of SA and SB in panax stems and leaves was 0.11% and 0.21%,respectively.
