The safety problem of environmental water has been widely concerned by people.Hepatitis a virus is a typical pathogen in water which can survive in water for a long time and it has a great impact on human health which already caused lots of disease outbreak in recent years.So HAV has raise people's attention in particularly.The content of HAV in environment water is too low to be detect and the detection of virus is very difficult,so the development in this area is relative slow.We need an efficient and reliable method to concentrate the HAV in water.People has committed on the concentration of virus in water for a long time.In this article,we have summarized the commonly used method of virus concentration and detection.According to the existing experience and methods,we created a new concentration methods which strong practicability.The content of virus was detected by RT-PCR to evaluate the effect of concentration method.The process of virus concentration was divided into two steps in this article.First of all,we concentrated 10 L or above HAV water sample to about1 L by polyether sulfone Ultra-filtration membrane and the Ultra-filtration membrane was backwashed to improve the recovery rate.The experimental data show that the recovery rate has up to 83%.Then the collected filtrate was further filtered by microfiltration membrane.The effects of filter membrane pore diameter and material,eluent,pH and time of eluting were compared.Finally,the result showed that 0.1?m mixed cellulose esters membrane showed highest HAV recovery rate which is up to 65%.We also concentrate 4 water samples in Dalian City including tap water and sea water with the method created in this paper.The result showed that all HAV recovery rate of practical water sample exceeds 40%.This method is of great significance to the concentrate HAV in environmental water.
