Elaeagnus angustifolia L.is a kind of pharmaceutical plants resources which can resistant toalkali and cold,it's distributed everywhere in Xinjiang.Elaeagnus angustifolia flavonoids can remove oxygen free radicals significantly,reduce cholesterol,control the inflammatory bioenzymes,it can accelerate wound healing and relieve pain.In this paper,Angustifolia was used as the research object,which mainly focused on the extraction and purification of flavonoids from Angustifolia.The application of flavone was preliminarily studied.It provides a theoretical basis for further improving the application value of Elaeagnus angustifolia.The main conclusions in this paper are as follows:The technology of flavonoids extracted from Angustifolia by traditional solvent extraction method,Ultrasonic-assisted extraction method and Microwave-assisted extraction method was studied.The extraction process was optimized by single factor and response surface methods.Comparing the three extraction methods,the experimental results are as follows:The ultrasonic-assisted extraction has the highest content,followed by microwave-assisted extraction,and the traditional solvent extraction method is the lowest.The optimized extraction technical conditions were as follows:extraction temperature was62.02?,solid-liquid ratio was 1:15.65,and ethanol concentration was 70.32%.The flavonoid content of Angustifolia reached the highest,the theoretical prediction value was 11.320 mg/g.The verification test results showed that the extraction temperature was 60°C,solid-liquid ratio was 1:15,the concentration of ethanol was 70%,and the extraction content reached 11.163 mg/g.Therefore,the ultrasonic-assisted extraction method was chosen as the extraction method of flavonoids from Angustifolia.By comparing the static adsorption capacity,adsorption and desorption rates of four kinds of macroporous resins with X-5,D101,AB-8 and NKA,it was determined that AB-8 macroporous resin is most suitable for the purification test of flavonoids from Agustifolia.Through the static single factor test on the purification of flavonoids from Agustifolia,it was determined that the pH of the test solution was 4,and the concentration of the test solution was 2 mg/mL as the optimal conditions.Through the dynamic single factor test of the purification of flavonoids of Agustifolia,the optimal conditions for volume of absorbing solution were determined:4BV,the flow rate of absorbing solution:2BV/h,volume of eluant:4BV,the flow rate of eluent:2BV/h.The purity of flavonoids of Angustifolia is measured under the condition of optimum purification technical of 45.21%and the purity was 15.54%before purification.Antioxidant performance of extracts of Angustifolia flavonoids and vitamin C were investigated with total reduction ability,hydroxyl free radical system,superoxide anion free radical system.The scavenging rates of the present extract on hydroxyl radical and radical superoxide were 85%and 88.23%at concentrations of 4 mg/mL,that the antioxidations from extracts were better than vitamin C.The stability of flavonoids extracted was studied,the content of Angustifolia flavonoids had decreased for 10.08%with light illumination after 7 days,and that absorbancy had significant change in 80?for 10h;the Angustifolia flavonoids were stable at pH=6-7.In addition,Na+and K+exhibit little effect on the stability of Angustifolia flavonoids,however,Cu2+and Fe3+have obvious influence on the stability of flavonoids,avoid contact with Cu2+and Fe3+.The results of the study showed that the optimum technological parameters for the preparation of the Angustifolia flavone oral liquid were as follows:the extract of Angustifolia flavone was 5%,xylitol was6%,maltose was 10%and citric acid was 0.3%.The oral liquid under this condition is suitable for sweet and sour,with a light fragrance,suitable color,and meets the taste of the public.
