The Zheng Benqing Tea is produced by hengdu Traditional hinese Medicine Health are Technology co.,LTD,which made up of edible hinese herbs.The formulation named “Detoxification drug composition and its preparation method and application” was patented by the state.In order to develop it into a health food with the characteristics of hinese medicine,this project had carried out preliminary research on functional test of Zheng Benqing Tea.Moreover,the scientific production process and quality control methods were established.In addition,the advanced high resolution mass spectrometry technique was used to identify potential functional substances.Through these methods,it laid a foundation for further exploring the effect and mechanism,the relationship between pharmacodynamic substances and the declaration of health food(This project originated from a project of Science and technology innovation seedling project and Chengdu Huimin projects of science and technology of sichuan province).The details are as follows:1.Methods for quality control and standard drafting of the Zheng Benqing Tea(1)The HPL wavelength switching method was established to determine the content of gallic acid,chlorogenic acid and geniposide of Zheng Benqing Tea at the same time.The experimental results showed that the established method can separate the three substances from the untested components well,and the method is stable and reliable which is easy to operate.(2)The processing parameters of Zheng Benqing Tea production were preferred.Box-Behnken experimental design with OD value was employed to optimize three main parameters: drying time,drying temperature,crushing size based on single factor tests taking contents of water soluble extract,gallic acid,chlorogenic acid,gardenoside as evaluation index.As a result,the optimal processing conditions were determined.(3)A fingerprint analysis method for the quality control of Zheng Benqing Tea? was established.Using high performance liquid-ultraviolet detector,several samples were measured.Nineteen common peaks within 105 min were well separated,four of which were identify through reference substances and some of which had been assigned.Ten batches of Zheng Benqing Tea samples were analyzed by Similarity evaluation system for chromatographic fingerprint of traditional hinese medicine(2004 A).All of the similarities were above 0.95,showing that the Zheng Benqing Tea's quality was in good consistency and high stability.So,this method can be effectively used in the quality control of the Zheng Benqing Tea.(4)The methods of morphological identification and TL identification were established.Water,ash,heavy metal,microorganism et al were examined.Finally,the enterprise standard of Zheng Benqing Tea was drawn up,which provided the relevant basis for the quality control.2.Defecation and lipid-lowering pharmacodynamics of the Zheng Benqing TeaDefecation pharmacodynamics of Zheng Benqing Tea was studied by constipation model with compound diphenoxylate through observing the defecation condition and the movement of small intestine of mice.As a result,Zheng Benqing Tea had a significant effect.Lipid-lowering pharmacodynamics of Zheng Benqing Tea was studied by hyperlipidemia model with hyperlipidemia feed through determining and analyzing the TG,T,HDL,LDL-in mice serum.As a result,Zheng Benqing Tea was no obvious effect of reducing blood lipid.3.Chemical profiling of Zheng Benqing Tea by ultra high performance liquid chromatography coupled with hybrid quadrupole orbitrap mass spectrometryAn ultra high performance liquid chromatography coupled with hybrid quadrupole orbitrap mass spectrometry method was developed to systematically analyze chemical constituents of Zheng Benqing Tea's aqueous solution.onstituents were identified by HRMS in both positive and negative ion mode using full scan and two stage threshold triggered mass modes.A total of 39 compounds,including 4 amino acids,5 flavonoids,8 triterpenoid saponins,13 organic acids,6iridoid glycosides,2 single terpenoids,1crocus pigments were identified byanalyzing their high resolution mass spectral data comparing with reference standards and open database even a large number of literature.This study provides a material basis for further study on the efficacy and pharmacodynamics of Zheng Benqing Tea,on the other hand,it demonstrates that hybrid quadrupole orbitrap mass spectrometry is a powerful tool in structural identification of unknown compounds in complicated hinese herbal compound prescription,which provides reference for establishing a more scientific and reasonable quality control method of prescription in future.
