In this paper,Soybean source of sterols were used to study the High fat and high cholesterol diet hamsters,we studied the efficacy and mechanism of soybean steroids in lipid-lowering and cholesterol-lowering in the body.The situation of the changes of intestinal flora under the condition of high-fat diet is preliminary described and Soybean source of sterols regulate Hamster disorders of intestinal bacteria caused by high fat and cholesterol diet.It is expected to provide more scientific theoretical basis for the study of cholesterol-lowering mechanism of intestinal flora,for the development and utilization of the Cholesterol-lowering functional factor and foods of intestinal microflora to lay a better foundation.The experiment is divided into 5 groups in total,Normal control group,High-fat group hypercholesterolemic diet-induced,the other three groups received dietary interventions with soybean source of sterols,drug concentrations were 1 ‰?5‰?10‰,and the experiment was conducted for 6 weeks.Blood was collected at weeks 0?3.6 respectively,The kit measured the levels of(TC),(TG)and(HDL-C)in hamster serum,the excretion of steroids in feces and cholesterol content in the live were detected by gas chromatography.Real-time quantitative polymerase chain reaction(qPCR)analyzed genes that regulate cholesterol metabolism of small intestine and liver.Denaturation gradient gel electrophoresis(DGGE)was used to analyze the trend of intestinal flora in hamsters.results show soybean source of sterols group can significantly reduce the levels of serum TC,TG and the Non-HDL-C,Hamster small intestine,liver cholesterol-regulating genes such as SREBP-2,LXRa,HMG-CoA-R,LDL-R mRNA downregulation affected by high-fat diet.The gene expression of down-regulated gene was significantly increased after soybean sterol intervention(P<0.05).Other genes such as NPCILI,ABCG5/ABCG8 mRNA were up-regulated or not affected.Soybean steroids can also promote steroid excretion in feces while blocking the absorption of cholesterol,and significantly reduce the cholesterol content in the liver(P<0.01).Soybean steroids could also promote the excretion of sterol in the feces while preventing the cholesterol absorption,and significantly reduce the cholesterol content in the liver(P<0.01)DGGE results showed that high-fat and high cholesterol diet increased the diversity of intestinal flora,After giving soybean source of sterols the uniformity of intestinal flora was significantly increased,Intestinal environment tends to balance,it is good for beneficial bacteria to colonization and Inhibit the proliferation of harmful bacteria.Summary of the paper,to some extent,high-fat diet reducing the number of predominant bacteria(Bacteroides sp?Ruminococcus.sp),increasing the number of harmful bacteria(Clostridium sp?Oscillibacter sp.),in hamster intestinal,causing mild inflammation of the intestine.The addition of soybean source of sterols improves the composition of microbial Intestinal Structures.Indirectly affecting the synthesis of cholesterol,absorption and discharge,so as to achieve the effect of hypolipidemic.
