| Polysaccharides were extracted and purified from abalone gonad,and hepatoprotective activity,antifatigue activity and antithrombotic activity were investigated.Three polysaccharide fractions,named AGP-11,AGP-12 and AGP-13,were obtained from gonad of abalone Haliotis Discus Hannai Ino by protease hydrolysis and ethanol precipitation and followed by further purifications with DEAE-52 anion exchange chromatography.The protein content of the three components were 9.36%,7.06%,8.68%,The Polysaccharide content were 52.29%,57.24%,57.52%,sulfate radical content were 0.4%10.22%,9.99%,alduronic acid content were 7.31%,12.59%,14.85%.The molecular masses of AGP-11,AGP-12 and AGP-13 were 8 kDa,22 kDa,64 kDa.AGP-11,AGP-12 and AGP-13 shared the same monosaccharide as rhamnose(Rha),mannose(Man),galacitiol(Gla)and glucose(Glc).The ratio for AGP-11 was Rha:Man:Gla:Glc=5.8:8.3:1.0:8.0,AGP-12 was Man:Rha:Glc=1.0:10.0:6.4.AGP-13 was Rha:Man:Glc=1.1:1.3:1.Experimental results indicated that AGP-13 could protect mice liver from acute injured by CCl4.AGP-13 could decrease the content of serum GPT(P<0.01),GOT(P<0.05)and hepatic tissue MDA.And it could also increase the activity of SOD(P<0.01)and GSH-PX.In study on the antifatigue activity of CAGP,we found CAGP could prolong the swimming time of loaded mice(P<0.01),increase the content of hepatic glycogen,hemoglobin and the activity of serum lactate dehydrogenase(P<0.01).CAGP could also increase the content of musule glycogen(P<0.05)and decrease the content of serum lactate.And we also found that polysaccharides which were extracted from abalone gonad had the potential effect of thrombolytic therapy.Therefor we had a conclusion that polysaccharides extracted from abalone gonad have hepatoprotective effect,antifatigue effect and potential effect of antithrombotic activity. |
