With the accelerated pace of life,people are bearing more and more stresses from study and work.Accumulating evidence indicated that chronic stress is an important causal factor for the development of depression.Depression is one of the most severe psychiatric disorder worldwide.Over 10% of the global population is suffering from mood,anxiety disorders.Sesame lignans,including sesamin,sesamol and sesamolin,are the major bioactive components in sesame seed,sesame oil,and sesame cake.Recently,the health food industry develop rapidly and have the very broad market prospect.The present research is aimed at uncovering the effects of sesame lignans extract from sesame cake on chronic stress-induced depression-like behavior disorders,and developing a new healthy sesame lignans extract food pattern.The major aspects are as following:(1)The content of sesame lignans from sesame meal was applied as the index of evaluation to investigate the extraction process.The results show that the best process parameters of ultrasonic-assisted extraction of sesame lignans,were as follows: 80% ethanol,55 °C,2 hours,solid-liquid ratio 1:8,the optimum yield of sesame lignans was 0.11% under this condition.The content of total of sesame lignans,sesamin,sesamol and sesamolin is4520.4 mg/100 g,3298.1 mg/100 g,969.2 mg/100 g,302.7 mg/100 g,respectively.the extract containing sesame lignans can be used to treat mice.(2)The most acceptable chronic stress model for screening anti-depressants is unpredictable mild stress(CUMS)rodent model.We studied the effects of sesame lignans extract supplementation on CUMS induced depression mice model by characterizing the effects on mice anxiety and depression behavioral tests and examining the level of neurotransmitterin brain and plasma.The results indicated that administration of sesame lignans extract significantly alleviated the CUMS-induced depression-like behavior,including sucrose preference reduction,locomotor activity decline,and hypomotility.Sesame lignans extract treatment increased the concentrations of serotonin and norepinephrine in the hippocampus and prefrontal cortex of CUMS mice.Moreover,sesame lignans extract treatment also restored the morphology of the dendrites and increased postsynaptic densityprotein of CUMS mice.Thus,sesame lignans extract improve depression-like behavior disorders and relieve stress in depression-like behavior,anxietybehavior,memory,neurotransmitter and postsynaptic density proteins.The research provides the foundation for developing a new health anti-depressant food pattern.(3)Based on the functional outcome of the previous study,a new chewable sesame lignans tablet with good texture,was developed.The results showed that the optimum recipe of the sesame lignans chewable tablets,consist of 0.035%-0.35% sesame lignans,50%-70%sweet agent,1%-5.5% sour agent and 0.4%-1% lubricants.And the optimum recipe has the ability toexerts antidepressant-like effects.Samples 5 chewable sesame lignans tablet was optimized within the scope of the above formula,which comprises 0.18%sesame lignans from extract,60% sweet agent,3% sour agent,0.6% lubricants.In the present study,we demonstrated that the content of total of sesame lignans and other components through ultra-wave soaking under the certain conditions.Moreover,the results revealed that sesame lignans extract treatment ameliorated the depression-like behavior induced by chronic stress in a mouse model,including anhedonia,locomotor activity decline,and hypomotility.We developed a new kind of chewable tablet containing sesame lignans,which possesses good taste as well as antidepressant-like effects.Therefore,the current research could provide technical support and theory basis for the development of antidepressant food in the future.
