Objective To determin the prescription and preparation process of Wujiaqi drinks,then establish its quality standarads and study its stability,function of soothing nerves and nourishing brain,acute toxicity.Methods?1?Using water decoction prepared the normal juice.The recipe screening and preparation process were studied by singlefactor text and orthogonal test.Using natural clarification,centrifugal separation,heart coagulation,freeze clarification,microporous membrane and other methods were studied to clarify the process.?3?Producing three batches of Wujiaqi drinks,then observed the oral traits,density,pH value,soluble solids content and protein content,TLC was used to identify Schisandrae and Acanthopanax senticosus in this preparation.HPLC was used to determine the contents of schisandrin and syringin.Phenol sulfuric acid method was used to determine the contents of lycium barbarum polysaccharide.?4?Using accelerated test andlong-term test to measure internal control targets,then study its stability.?5?Through establishing the sleep disordermodel and pregnant mouse model,to determine the effect of drinks on sedation and improve learning and memory in mice and pregnant mice,then we have determined 5-HT,DA,Ach and Ache in the brains of mice and pregnant mice.?6?Through the median lethal dose in mice and the maximum tolerated dose trial to study the Wujiaqi drinks oral solution acute toxicity indicators.Results?1?The formula of the drink was that Acanthopanax juice:Schisandra juice:wolfberry juice:White Chrysanthemum juice:rock sugar solution:white sugar solution:drinking water?volume ratio?was 2:1:2:3:1:4:16.Clarification process test ofdrinks was used centrifugal clarification andmicroporous membrane method.?2?TheWujiaqi drinks was orange translucent liquid,uniform texture,no impurities,no suspended matter,no layered precipitation.Density of drinks was 1.040±0.007 mg·mL-1.pH value of drinks was 3.46±0.05.Soluble solids content was 8.5±0.3%.Protein content in per 100 mL drinks was 46.10±0.25 mL.The contents of schisandrin in per 100 mL drinks was 0.6645±0.0032 mg.The contents of syringin in per 100 mL drinks was0.160±0.023 mg.The contents of polysaccharide was 5.64±0.17 mg?mL-1.?3?To high temperature is relatively stable.Sample test were at high temperature for a long timeto stay6 months and room temperature for a long time to stay 9 months,oral traits,density,pH value,soluble solids content were not changed significantly.?4?In the sedative and tranquilizing study of Wujiayu drinks,as compared with the model group,the DA content in the brains of mice and pregnant rats increased,the content of 5-HT decreased in the brains of mice and pregnant rats,in the study of improving learning and memory of Wujiayu drinks,the Ach content in the brain of mice and pregnant mice increased,and the AchE content decreased.?5?The results of lethal dose test indicated that all dose groups no mortality and no adverse reactions.The results of MTD showed that the maximum tolerated dose in mice to clinical 60 kg person per kilogram body weight daily dose of 240times.Conclusion The subject is suitable for the preparation of Wujiaqi drinks,the process is reasonable and simple,and the operability is strong,the quality control method is simple,the precision,stability,reproducibility is good,and the product quality can beeffectively controlled,the functional research proves that drinks have a certain effect on improving sleep disorders and enhancing learning and memory,acute toxicity tests show that maximum tolerated dose in mice is 240 times higher than that of humans,which proves that the Wujiaqi drinks is safe and non-toxic.
