The fermented powde of Ganoderma Lucidum was used as material,and the extraction conditions of triterpenoidswere studied and its structure were identificated.The solid-state fermentation conditions of Ganoderma Lucidum were optimized.The results were as follows.1.Thetriterpenoids of Ganoderma lucidum were extracted by water bath reflux and ultrasonic-assisted extraction.The results showed that the water bath reflux was the better method.and the extraction condition were as follows:the extraction solvent was 95%ethanol,soaking 80min,the ratio of solid and liquid was 1:30(g/mL),refluxing at 85 ? for 3 hours and extract 3 times,,the yield of triterpenoids was 0.6760 mg/g.The results provided theoretical basis for online monitoring of fermentation products from Ganoderma lucidum.2.Through single-factor experiments,it was found that the growth of Ganoderma lucidum mycelium and the production of functional components were effected by Ganoderma lucidum strains,fermentation bases,fermentation temperature,fermentation time and inoculation amount.The results showed that the best fermentation process was as follows:550g corn as the base material,511096(bonsai No.3)of liquid bacteria species 35mL,fermentation for 15 days at 28? the yield of triterpenoids was 1.6100 mg/g and the content of polysaccharide was 7.1087%.3.The triterpenoids were purified by silica gel column chromatography.The purified samples were identified qualitatively by thin layer chromatography.The samples were separated by different developing systems and the best developing system was the volume ratio of toluene,ethyl acetateand acetic acid was 12:4:0.5,the chloroform and methanol were used as the eluent and the optimum ratio was90:10,and chloroform:methanol=20:1.4.The mobile phase condition of high performance liquid chromatography was optimized.The 2%acetic acid solution and acetonitrile were used as the mobile phase and the best ratio was 70:30.the structure of triterpenoids from Ganoderma lucidum was analyzed by ifrared and ultraviolet spectrum respectively and the results showed that the structure of triterpenoids from Ganoderma lucidum were a kind of tetracyclic triterpenoid compound containing a carbonyl group.
