Recent years, people have paid more attention to the research of Ganoderma lucidum because of the increasing artificial breeding of Ganoderma lucidum. However, some active ingredients from Ganoderma lucidum are not explored and studied completely, resulting in a waste of Ganoderma lucidum resources. This thesis mainly illustrated the extraction, purification, analysis and antioxidation of triterpenoids from Ganoderma lucidum in Guan County, Shandong province.This thesis compared different extraction methods to determine the optimal experimental conditions by response surface method and purified the extract by using macroporous resin. After selection of macroporous resin, static and dynamic absorption kinetics experiment, elution requirements were determined. Then analysing the Ganoderma lucidum triterpenoids by using HPLC-MS identification method. Finally, antioxidations of different extracts were compared. Results as follows:1. Ultra high pressure method was firstly adopted in Ganoderma lucidum triterpenoids extraction, which was compared with traditional solvent extraction and ultrasonic-assisted extraction. After optimizing the experimental conditions, the evidence showed that Ganoderma lucidum triterpenoids by ultra high pressure method owned the largest amount with the minimum time. The optimal technological conditons are:extraction time 5min, ethanol concentrations 96%, material-solvent ratio 1:26 g/mL, pressure 295 MPa, temperature 40℃.The content of total Ganoderma lucidum triterpenoids is predicted to be around 1.3200%.2. Purifying the previous extract from ultra high pressure processing by using macroporous resin. Determining the best condition of purification by selection of macroporous resin, static and dynamic absorption kinetics experiment:AB-8 macroporous resin, sample amount 30mL, speed 2BV/h, pH value of sample 4, elution order as follows:distilled water,15% ethanol,35% ethanol,95% ethanol.3. Acetonitrile-water was chosen to be the mobile phase by analysing the high performance liquid chromatography separation chromatographic conditions of Ganoderma lucidum triterpenoids. Through the liquid chromatography-mass spectrometry analysis, Ganoderic acids were preliminary supposed to be involved in the extractions.4. Antioxidant activities were investigated by comparing different extractions. The antioxidant activities of these extracts were analysed against Vc and BHT by using DPPH radical scavenging activities, hydroxyl radical scavenging activities, ferric reducing antioxidant power and superoxide anion radical scavenging. The results showed that ultra high pressure-assisted extracts exhibited more remarkable than other extracts. The larger amounts of triterpenoids owned, the better antionxidant resistence behaved.
