Our country is the second largest wine grape growing area in the world,and the reuse of winemaking grape pomace is the hot spot in the recent research on functional food processing.Xin Jiang winemaking cabernet sauvignon skin residue as material in this project,optimization within established the theoretical basis of energy density,the response surface method was used to optimize the extraction process of procyanidins and grape seed oil.The main active ingredient in grape skin residue is anthocyanin,resveratrol and grape seed oil,preparation of procyanidin,resveratrol and grape seed oil complex.Using HPLC method,GC-MS method,to detect the contents of procyanidin,resveratrol and fatty acid were analyzed.Establish a murine hyperlipemia model,using 2󫎾 factorial experiment,discussed the interaction between low and high doses of these substances.The research of this subject has great economic and social benefits,which is of great significance to the sustainable development of wine industry.The conclusion as follow:?1?The extraction process of the procyanidins was established by the combination of the cohesive energy density and the response surface method.The results showed:extract the solvent as methanol:1%hydrochloric acid?95:5?,material liquid as 1:20,60?water bath heating 100min,parallel extracts2times,the average content of the procyanidin was 49.39mg穏-1in this extraction condition.Higher than the average in the literature.?2?The extraction process of grape seed oil in winemake grape pomace was established by the cohesive energy density and response surface method.The results showed:extract the solvent as N-hexane:cyclohexane?11.6:88.4?,material liquid as 1:8,80?water bath heating 120 min,55?rotary evaporation to without solvent,parallel extracted 2 times.The oil rate was 14.87%.The GC-MS test results in four aliphatic acids in grape seed oil,which is linoleic acid?52.72%?,followed by oleic acid?32.92%?,palmitate?7.41%?,stearic acid?6.95%?,the oleic acid and oleic acid unsaturated fatty acids were 85.64%of the aliphatic acids.The degree of unsaturation of grape seed oil was indicated.?3?Established 2󫎾 murine hypolipidemic factorial experiment,the extract of the procyanidin was set up as 100mg/(kg.d-1)?200mg/(kg.d-1),resveratrol as 50mg/(kg.d-1)?100mg/(kg.d-1),grape seed oil as 20mL/(kg.d-1)?30mL/(kg.d-1),to be low and high dose groups for the experiment.Through SPSS software analysis,the optimum group of hypolipidemic was selected the original anthocyanin extracts 200mg/(kg.d-1),resveratrol 50mg/(kg.d-1)and grapeseed oil20 mL/(kg.d-1).?4?HPLC method to determined the content of procyanidin and resveratrol in procyanidinol resveratrol and grape seed oil complex,determined the correct dose of procyanidin and resveratrol in murine hypolipidemic factorial experiment.The content of the procyanidins in the compound was obtained26.5%?n=3,RSD%=0.59%?;the content of resveratrol in the compound was obtained 23.8%?n=3,RSD%=0.29%?.
