Objective Nano-selenium of different sizes were prepared.The particle size,morphology and selenium content were determinated and the antioxidant capacity in vitro,the hepatoprotective and hypoglycemic activities in mice were investigated of different sizes of nano-selenium.Methods Using ultrasonic-assisted Vc reduction to prepare different sizes nano-selenium of selenite with PEG2000 as template and selenite as selenium source.Taking the size of nano-selenium as the index and the different sizes nano-selenium were preparated by orthogonal experiment.Different sizes nano-selenium were characterized by particle size analyzer,dual-wavelength method of colloidal solution and scanning electron microscope.The selenium content was determined by o-phenylenediamine ultraviolet spectrophotometry of nano-selenium.The effect on scavenging hydroxyl and DPPH free radicalsradicals were determined by using UV spectrophotometric of different size nano-selenium.The reducing ability was determined by the Prussian blue method of the different sizes nano-selenium.SPF Kunming mice were divided into normal group,negative group,positive group,small,medium and large size nano-selenium group.Mice were induced acute liver injury by Carbonation,intragastric administration the mice for 14 days except for the normal group with different size nano-selenium.Serum biochemical indicators were measured to evaluate the degree of liver damage,HE stain of liver tissues were pathological observated.Mice were induced diabetic by alloxan and were randomly divided into normol group,negative group,positive group,small,medium and large size.Intragastric administration the mice for 21 days except for the normal group with different size nano-selenium.Serum biochemical indicators were measured to evaluate the degree of diabetic level,HE stain of liver and kidney tissues were opathological bservated in mice.The effects on hepatoprotective and hypoglycemic activities were evaluated of different sizes nano-selenium.Results The preparation of small-size nano-selenium was 0.5 h at 30?,the ratio of Vc to selenite was 3:1,the concentration of PEG2000 was 3.0 mg·mL-1.The particle size at about 60nm,80 nm,100 nm were as small,medium and large size nano-selenium respectively according to the experiment.Scanning electron microscopy showed that the different sizes nano-seleniums were spherical and uniform.The selenium content in small,medium and large sizes were130.89,221.67,and 392.94?g·mL-1 of nano-selenium,respectively.The anti-oxidation results of nano-selenium showed that the IC50 values were 0.075 mg and 0.122 mg,respectively;of Vc and small-size nano-selenium scavenging hydroxyl radicals;scavenging DPPH free radicals were 0.027,0.025,0.035 and 0.038 mg,of;Vc and small,medium and large-size nano-selenium.The reducing ability was Vc>small size>medium size>large size nano-selenium.The small and medium sizes nano-selenium had a good protective effect on acute liver injury induced by CCl4 in mice,which could reduce the liver weight index of mice and the ALP activity in mice serum.The levels of ALT,AST in serum,white ball and bilirubin?P<0.01?increased the levels of TP and HDL-C.The pathological observation of liver tissue showed that small and medium-sized nano-selenium could protect liver cells and the small size nano>medium size>large size nano selenium.The results of hypoglycemia in mice indicate that nano-selenium can effectively increase the body weight of diabetic mice,and there was no significant difference between the first week and the second week of diabetes in the treatment of diabetes?P>0.05?.After the first week of administration,the blood glucose of the mice was significantly decreased which showed significant difference?P<0.01?between the small and medium-sized nano-selenium groups and the negative group in the second week and the third week.The small size of could increase the serum levels of TG,TP and TC,and than negative group?P<0.05?.The small nano-selenium can significantly reduce the UREA content than negative group in serum?P<0.01?;the HE staining of liver and kidney tissues,found that diabetes were not damage of the liver,but the kidney tissue of the negative group were severely damaged.The damage of renal cells in different sizes nano-selenium group and positive group were little.Conclusions The preparation method was simple and easy,and the prepared nano-selenium had a concentrated particle size,stable morphology and uniform dispersion.Different sizes nano-selenium had strong anti-oxidation effect in vitro,and the anti-oxidation ability was negatively correlated with the nano-selenium particle size.In addition,nano-selenium had a good protective and therapeutic effect on acute liver injury and diabetes.The smaller the particle size of nano-selenium,the better the effect.This provides an experimental and theoretical basis for the future research of biological activity.
