This thesis consists of three chapters.The first chapter mainly reviewed the biological activity of xanthone derivatives.The second chapter reported the purification and antioxidant activity of total xanthones in the roots of Swertia kingii Hook.f..The third chapter reported the study on quality standard of Shi Wei Hei Bing Pian Wan.Chapter 1 mainly searched through scientific search engines such as SciFinder,Google Scholar,Web of Science and MEDLINE.Through reviewing domestic and foreign literatures,the current research status of biological activities of xanthone derivatives was reviewed.Chapter 2 consists of two parts.The first part examined the refining process of total xanthones in the roots of Swertia kingii Hook.F..The D-101 macroporous resin and water sedimentation-acidic precipitation method were used to purify the total xanthones in the roots of Swertia kingii Hook F.,and the water sedimentation-acidic precipitation process was optimized.The best process was used to isolate and purify total xanthones.The best refining process is as follows: the concentration of the drug solution is 0.1 g crude drug/mL ethanol,and the amount of ultrapure water is twice the total volume of the drug solution,standing,centrifugation(9000 rpm,8 min),after obtaining the water-sedimentation product,add 10 times of HCl solution with a pH of 3.0,ultrasonically disperse for 5 min,perform acidic-precipitation test,let it stand for 1 h,and centrifuge(9000 rpm,8 min)to obtain a precipitate.The precipitate was washed with distilled water until neutral,and dried under vacuum to obtain total xanthones.In the second part,a certain amount of total xanthones is prepared by the preferred purification process,and the antioxidant activity of total xanthones was evaluated by DPPH method,FRAP method and hydroxyl radical method.Chapter 3 consists of five parts.The first part is the microscopic identification of Shi Wei Hei Bing Pian Wan.Microscopic identification identified the characteristics of nine kinds of medicine except bear bile.The second part is the physical and chemical identification of Shi Wei Hei Bing Pian Wan,in which the physical and chemical identification of bright salt,Herpetospermum pedunculosum and Cinnamomum cassia Presl is better.The third part is thin-layer chromatographic identification of Shi Wei Hei Bing Pian Wan.The TLC identification of Punica granatum L.,Terminalia chebula Retz.,Piper longum L.,Herpetospermum pedunculosum,and Holarrhena antidysenteriaca Wall.ex A.DC.is better.The fourth part is determination of gallic acid in Shi Wei Hei Bing Pian Wan.With a complete methodological study,the standard of gallic acid content is determined.The fifth part is the determination of heavy metals and harmful elements in Shi Wei Hei Bing Pian Wan.The contents of five heavy metals and harmful elements such as lead,copper,cadmium,mercury and arsenic are determined by ICP-MS and atomic fluorescence spectrophotometer.
