Inonotus baumii is one kind of rare medicinal fungus.Recent studies have shown that it contains a variety of active substances,which have effective therapy for many diseases including cancers,blood pressure,antioxidant and other pharmacological effects.However,the process of development and utilization of Inonotus baumii resources was slow,on account of the high growth requirements of the environment,long growth cycle,and insufficient research on chemical components.In this paper,the main product of Inonotus baumii fermentation broth was isolated and identified,and then the conditions of macroporous resin separation was optimized.Finally,a novel process of coupling fermentation with separation on Inonotus baumii was established.It provides the basis for development and utilization of Inonotus baumii.In this paper,the main product was isolated from the fermentation broth of Inonotus baumii,which was identified as a new styrylpyrone by spectroscopy.It was named phelligridin LA,which has stronger scavenging ability for DPPH-,O2-,OH-,ABTS+radicals and the total antioxidant activity was 23.99 U/mL.The optimal conditions for static adsorption were pH 5.19,the liquid-solid ratio 5.11,adsorption temperature 22.81?by RSM,in this condition adsorption rate was reached 97.11%.The optimal conditions for elution were70%ethanol.The adsorption process is in accordance with the Freundlich model,which is the spontaneous exothermic process,the hydrogen bond is the main adsorption force.The adsorption kinetics conforms to the second order kinetic equation,the adsorption rate is very fast.The adsorption mass transfer process mainly is influenced and controlled by the proliferating in the fluid film and inside the grains.The self-made fermentation reactor was ectopic coupling with the chromatographic column adsorption separation.The fermentation broth was pumped into resin chromatography at 1 mL/min on the seventh day,and eluted with70%ethanol at 5 mL/min,until the ninth day.The yield of phelligridin LA was increased 2.14times,and the purity of phelligridin LA was 97.53%by using this new process.It indicated that the new process have significant effect on enhacing production and separation of phelligridin LA.A new styrylpyrone compound phelligridin LA was isolated from Inonotus baumii for the first time in this subject,which is the main product of Inonotus baumii by fermentation and show a good antioxidant activity.A new process of coupling fermentation with separation on Inonotus baumii was established,which can enhance the yield of phelligridin LA and obtain comparatively pure phelligridin LA.This is undoubtedly provides a new separation of ideas for large-scale fermentation of Inonotus baumii and promotes the industrialization process of phelligridin LA.
