Chicken manure and corn leaf are agricultural resources with abundant nutrition and huge yield in agricultural production and animal breeding.Aerobic composting can solve the problem of environmental pollution and waste of resources caused by chicken manure and agricultural waste,in line with China's The development strategy of zero-growth of fertilizers and pesticides in major crops can also bring considerable economic benefits and is one of the effective ways to scientifically utilize resources.In the process of using it as a raw material for aerobic composting,the development of a sTable and efficient compound microbial pro-bacteria agent is a key factor in this technology.This paper aims to develop a comprehensive technology for the preparation of bio-organic fertilizers from chicken manure and corn glutinous rice using a composite microbial agent.First,high-efficiency lignin-degrading strains,cellulose-degrading strains,deodorizing strains,and other functional strains?protease strains,amylase strains?deposited in the laboratory were screened by methods such as enrichment culture,primary screening and rescreening.Scientific compatibility,using chicken manure and corn glutinous rice as raw materials,aerobic composting test by adding different kinds and proportion of microbial agents to determine the best combination of bacteria,and industrial production of fungicides,and finally use The production of composite microbial agents for large-scale composting applications and verification of its fertilizer efficiency.The main results are as follows:1.In this experiment,178 strains with cellulose-degrading ability were isolated and screened from vegeTable fields,corn stover and soil samples by heating enrichment of spores and Congo red medium transparent hydrolyzed ring.Quantitative determination of cellulase activity.Among them,a strain named T-7 had higher enzyme activity and the enzyme activity was 343.95 U/mL,which was identified as Bacillus subtilis.Using heating enrichment and initial screening of aniline blue hydrolysis circle,combined with quantitative determination of enzyme activity,87 strains with lignin-degrading ability were screened,and the fermentation broth was tested for lignin activity.One strain was named N3-8.The strain of had higher enzyme activity,wherein the lignin peroxidase activity was 2060.00 U/mL and the manganese peroxidase activity was 333.33 U/mL.The strain strain was identified as Bacillus licheniformis.18 strains with deodorizing effect were isolated and purified from fresh chicken manure and compost chicken manure by domestication and enrichment and plate scribing.After primary screening and rescreening,one of them was named C-9.The strain has the best deodorizing effect,and the removal rate of ammonia gas and hydrogen sulfide can reach 67.52%and 56.37%,and it is identified as Bacillus licheniformis.The enzyme activity of the protease strain and amylase strain preserved in the laboratory was determined,and the protease strain JA and the amylase strain DF-16 were selected as the strains for the subsequent research,and the protease activity and amylase activity were respectively determined.Both strains were468.56 U/mL and 397.72 U/mL,and both strains were Bacillus subtilis.2.The antagonistic test and coexistence test of each strain and the high temperature composting test of chicken manure and corn glutinous rice were carried out to determine the optimal combination of bacteria and bacteria.The composition of the fungicide was:cellulose degradation strain T-7+lignin The strain N3-8+deodorizing strain C-9+protease strain JA+amylase strain DF-16 was degraded.Using chicken manure and corn glutinous rice as raw materials,five different microbial pro-bacteria agents were added,and high-temperature composting tests were carried out under aerobic conditions.The organic matter,total nitrogen,total phosphorus,water-soluble organic carbon,seed germination index pH and heap temperature of the materials were measured at different stages of composting.The differences and trends of the above indicators in the application of five kinds of bacteriostatic agents were studied..The results showed that the addition of microbial agents could increase the temperature of the heap in the high temperature period,and the temperature of the heap increased significantly,and the duration of high temperature was significantly higher than that of the control group.The M6 was the most obvious effect on the heap;the experimental group and the control during the composting process.The pH of the group increased first,then decreased and then increased,and the pH value was maintained in the range of 7.09.0,which was beneficial to the growth and reproduction of microorganisms in the heap and accelerated the composting of the heap.The addition of microbial agents significantly degraded the organic matter.Among them,the effect of M6 is most obvious.During the composting process,the contents of total nitrogen and total phosphorus increase,and the content of water-soluble organic carbon decreases.The seed germination index of the heap can reach 80%or more in 21 days,which is in line with the national high temperature.The harmless requirements,the above indicators are most obvious with the addition of M6 bacteria agent:the addition of M6 bacteria agent increased the total nitrogen and total phosphorus content of the material by 27.9%,42.85%,organic matter,water-soluble organic carbon decreased by 51.93%56.1%,the temperature of the heap reached the high temperature period on the third day and lasted for 10 days,the maximum temperature was 58.9?,and the seed germination index of the heap finally reached 96%.The heap of the microbial agent M6 was added.Index compared to the control group vary significantly.3.Established the production process of various functional strains of composting composite bacteriaThe above strains were subjected to a 20 M3 fermenter liquid submerged fermentation spore-forming process and a post-treatment process,and spore powders of each functional strain were obtained.Taking cellulase-functional strains as an example,the optimal fermentation conditions are as follows:the medium formula of Bacillus sp.T-7 strain is sucrose 1.0%,peptone 1.0%,KH2PO4 0.1%,CaCl2·H2O0.05%,pH 7.0,The seedling age was 16 h,the inoculation amount was 4%,the culture temperature was 30?,and the rotation speed was 200 r·min-1 for 36 h.The biomass of the fermentation broth was 2.89×109 cfu·mL-1,and the spore formation rate was?95%.2 M3 seed tank culture conditions were:culture temperature 32?,optimal ventilation ratio 1:0.50.6,seed age 16 h,bacterial bacteria content 7.3×108cfu·mL-1.The fermentation conditions of 20 M3 fermenter were:4%seed,temperature 36?,ventilation 1:0.8,rotation speed 300 r·min-1,fermentation time 48h,and the final total biomass was 3.07×109 cfu·g-1.The centrifugal concentration conditions were as follows:the rotation speed was 7100 r·min-1,the feed flow rate was 0.6 T·h-1,the concentration ratio was 9 times,and the yield was 96%.The adsorbent is bran,and the optimum process parameters of the flash-drying post-treatment process are:air inlet temperature of 125?,outlet temperature of about80?,and feed volume of 90 kg per hour.The bacterial strain of the finally obtained Bacillus sp.T-7 strain was 2.97×10 10 cfu·g-1,and the water content was less than 5%.4.large-scale maturity application test and its fertilizer efficiency test.The spore powder microbial agents of each strain produced are proportioned into a composite microbial agent,and the composting system is connected to the inoculum of about 30 trillion viable bacteria per ton of material,and the whole nitrogen and the whole are carried out after 35 days.The indexes of phosphorus,organic matter and seed germination index showed that the total nitrogen and total phosphorus content increased by 37.9%and 43.85%,organic matter and water-soluble organic carbon decreased by 56.93%and 57.1%,and the seed germination index of the heap finally reached 98%.The organic fertilizer was applied to the pot experiment of wheat.The results showed that the plant was strong,the stems and leaves were thick and wide,the color was green,the fresh weight of the plants increased by 26.41%,the main root length increased by 55.56%,and the chlorophyll content increased by 87.41%.The nitrogen content increased by 253.27%.The total nitrogen content of the soil reached 0.536g/kg,an increase of 88.07%.In summary,this study obtained a functional strain that can efficiently promote high-temperature aerobic composting of chicken manure and corn glutinous rice,which was scientifically formulated into a composite microbial agent,and realized the industrial production of complex microbial agents.The application process is clear and the fertilizer effect is clear.
