Eucommia ulmoides seed oil is a high-quality woody oil resource.The oil is rich in unsaturated fatty acids,alpha linolenic acid content as high as 66%,and which is one of the plants with the hightest content of aerf-linolenic acid found so far.Eucommia ulmoides seed oil has the physiological functions of lowering blood lipid,anti-thrombosis,anti-inflammatory,inhibiting the occurrence and metastasis of cancer,preventing cardiovascular diseases,preventing diabetes and its complications such as physiological functions,is of great value of development and utilization.However,unsaturated fatty acids tend to react with oxygen during the processing and storaging of Eucommia ulmoides seed oil,resulting in the destruction of vitamins,essential fatty acids and other nutrients in Eucommia ulmoides seed oil,which has adverse effects on human health.In order to improve the stability of Eucommia ulmoides seed oil,improve its low water solubility and poor chemical stability,this experiment prepared it into liposome,aiming to prolong its storage stability and achieve slow release in the intestine and improve bioavailability.This experiment provides reference for the research and development of functional oil carrier and theoretical basis and technical support for the industrial production of Eucommia ulmoides seed oil.1.Using ethanol injection ultrasonic method to prepare Eucommia ulmoides seed oil liposomes,tkake the encapsulation efficiency index,and based on single factor,through the Plackett-Burman experiment,the steepest slope test and the response surface experiment.The optimal preparation processes were:the mass ratio of soybean lecithin to?-sitosterol was 3.1:1,the mass ratio of soybean lecithin to Eucommia ulmoides seed oil was 4:1,the addition amount of Tween-80 was 20%,and the pH of PBS buffer was 6.9,the ultrasonic time was 13 min,and the ultrasonic power was 180W.Under the optimized conditions,the appearance of Eucommia ulmoides seed oil liposome is smooth and complete,spherical,with encapsulation efficiency of(75.26±1.58%),average particle size was(171.85±1.41 nm),PDI was(0.219±0.042).Zeta potential was(-19.3±1.56 m V),and has a good particle size distribution and stability.2.The lyophilization process of Eucommia ulmoides seed oil liposome was studied,and the appearance,redispersibility and encapsulation efficiency of the samples after refreezing were investigated.The results showed that the pre-freezing temperature was-50?,the pre-freezing time was 12 h,the drying time was 36 h,the lyoprotectant was mannitol,and the mass ratio of mannitol to phospholipid was 8:1,the Eucommia ulmoides seed oil liposome had better freeze-drying effect,the encapsulation efficiency was(72.40±1.46%).TEM observation showed that Eucommia ulmoides seed oil freeze-dried liposome has a quasi-spherical shape and uniform particle size distribution.SEM observation showed that the lyophilized liposome of Eucommia ulmoides seed oil had smooth surface and irregular shape.The particle size,PDI and Zeta potentials of Eucommia ulmoides seed oil freeze-dried liposome were(389.67±4.81 nm),(0.255±0.013),(-22.62±1.66 m V)respectively,indicating that the freeze-dried Eucommia ulmoides seed oil lyophilized liposome has a better particle size distribution and stability.The storage stability experiment showed that the Eucommia ulmoides seed oil freeze-dried liposome has a better storage stability at 4?and 25?.3.Explored the release mechanism of Eucommia ulmoides seed oil liposome,and found that the first-order kinetic equation was used to fit the release coefficient of the Eucommia ulmoides seed oil,and the first-order fitting equation of Eucommia ulmoides seed oil liposome was Q=54.930(1-exp(-0.4521t)),R~2=0.9046,the release in the Eucommia ulmoides seed oil liposome was mainly determined by the diffusion control mechanism,and the release was dominated by the Fick diffusion mechanism.In vitro simulated digestion experiment showed that the FFA release rate from Eucommia ulmoides seed oil liposome(69.42±1.86%)was significantly higher than that of the control group(57.84±2.44%).In the stage of simulating saliva and gastric juice,the system of Eucommia ulmoides seed liposome accumulates,the particle size increased,and the Zeta potential decreased.After digestion by simulated intestinal juice,the average particle size of the system suddenly decreased,and the Zeta potential increased sharply.The bioavailability of Eucommia ulmoides seed oil liposome was(63.48±2.57%),which was significantly higher than that of the control group(51.96±2.43%)(P<0.05).
