Eucommia ulmoides Oliver(EUO,also known as Duzhong in Chinese)is the unique living deciduous woody oil tree and precious medicinal herbs in China.Eucommia ulmoides seed oil is an oil produced from the seeds of Eucommia ulmoides,which is rich in polyunsaturated fatty acids and nutritional value.In the present study,the Eucommia ulmoides seed oil nanoemulsion was prepared by dynamic high-pressure microfluidization(DHPM).The main results are as follows:1.The formulation of Eucommia ulmoides seed oil microemulsion was optimized by water titration and conventional stirring method.According to the size of the microemulsion area and the proportion of each component,Tween 80 and anhydrous ethanol were selected as composite surfactants in the ratio of 2:1.The above composite surfactant with better emulsification can be directly used to prepare Eucommia ulmoides seed oil nanoemulsion by DHPM.According to the single factor and response surface optimization analysis,when the homogenization pressure was 117 MPa,the volume fraction of Eucommia ulmoides seed oil was 7.9%,and the surfactant concentration was 0.12 g/mL,the average particle size of the nanoemulsion was the smallest and the [-]?-potential was the largest.The nanoemulsion prepared under this condition had strong stability and uniform distribution,and the characters of the nanoemulsion formed under the optimal preparation conditions would be evaluated in the following chapters.2.The quality and characteristics of Eucommia ulmoides seed oil nanoemulsion were characterized.It could be seen that the appearance of nanoemulsion was a translucent milky white liquid,the type was oil-in-water(O/W).The average particle size was 187.63 nm,PSD showed monomodal size distribution and the range was 10-1000 nm,[-]?-potential was 19.07 mV and the distribution was uniform.Microstructure,rheological properties,and physical stability analysis showed that the newly prepared Eucommia ulmoides seed oil nanoemulsion particles were homogeneous,with excellent kinetic stability,and there was no precipitation,floating lipid and polymerization.Eucommia ulmoides seed oil extracted from the nanoemulsion and the oil all contained eight kinds of fatty acids,among which the contents of ?-linolenic acid,oleic acid,linoleic acid,and other primary fatty acids did not changesignificantly.Extending storage time and increasing ambient temperature will further reduce the stability of nanoemulsion.The increased rate of peroxide value of Eucommia ulmoides seed oil was faster than that of nanoemulsion,so the latter could significantly retard the oxidation of the oil.3.By analyzing the influence of environmental conditions on the stability of Eucommia ulmoides seed oil nanoemulsion,the stability of nanoemulsion under alkaline conditions(pH9-11)changed significantly more than that under acidic conditions(pH 3-7).At the same time,the lower ionic strength(0-300 mM NaCl)had no significant effect on the nanoemulsion,the heating temperature was relatively stable between 30-60?.When the temperature reached70?,the stability decreased significantly.In conclusion,the stability of Eucommia ulmoides seed oil nanoemulsion would be significantly affected under alkaline conditions(pH 9-11),higher ionic strength(>300 mM NaCl)and higher heat treatment temperature(>70?),and it could resist acidic pH conditions,which was more suitable for commercial development and utilization of acidic beverages.Additionally,Tween 80 could improve the stability of oil in the nanoemulsion delivery system.4.The Eucommia ulmoides seed oil nanoemulsion was added to the orange juice beverage as a functional material,and the shearing process was studied,it could be seen that when the shearing speed was 8000 r/min,the shearing time was 8 min,and the shearing temperature was 50?,the product had a better stabilization effect.The finished product not only showed milky orange in color and luster but also had the proper aroma and taste of orange juice.The most critical is to attach the unique lipid fragrance of Eucommia ulmoides seed oil with uniform texture,no oil floating,and stratification.Physical and chemical indicators,trace element content,and microbial indicators were within the scope of China standards.It should be stored,transported,and sold at a temperature above zero or average temperatures.
