Ganoderma lucidum is a traditional and valuable traditional Chinese medicine in China.Ganoderma lucidum contains many kinds of active ingredients.Among them,the pharmacological research on Ganoderma lucidum polysaccharide(GLP)and Ganoderma lucidum triterpene(GLT)is the most extensive.GLP have anti-cancer,hypoglycemic,intestinal function,and atherosclerosis treatment effects.GLT have protective medicinal value such as liver protection,anti-virus,and prostate disease treatment.It mainly contains GLP and GLT and other active ingredients.As an emerging technology,ultra-high pressure technology,because of its high extraction rate,short extraction time,and can maintain the same biological activity,has a great role in the field of traditional Chinese medicine,so it has received extensive attention in extraction.At present,there are few reports on ultrahigh pressure extraction of polysaccharides and triterpenes.Ultrahigh pressure technology is used to extract GLP and GLT in Ganoderma lucidum spore powder,which provides a new method for the development and application of comprehensive development of Ganoderma lucidum products.Ideas.The research mainly examines the influence of the extraction rate of GLP and GLT by changing the process operation parameters,such as extraction pressure,holding time,liquid-solid ratio and so on.Based on the influence of single factors,a response surface model is established to determine the optimal process parameters under experimental operating conditions.At the same time,in order to better study and describe the ultrahigh pressure extraction technology,the process of ultrahigh pressure extraction of GLP was analyzed using a kinetic model.Through the establishment and verification of the kinetic model,it was confirmed that the model could describe the ultrahigh pressure better the process of extracting GLP.Provide a theoretical basis for the industrial application of the process technology.The specific content of the paper is as follows:For GLP,a standard curve for detecting polysaccharide content was established by the phenol sulfuric acid method,which was used to determine the extraction rate of GLP.Through the single factor experiment,the influence of various operating parameters on the extraction of GLP was investigated.The response surface analysis was used to establish the corresponding model.The optimal process operating parameters for ultrahigh pressure extraction of GLP were: pressure 360 MPa,liquid-solid ratio of 25 mL/g,pressure holding time of 4.5 min,yield of GLP 2.79 %.Using a scanning electron microscope,from a microscopic perspective,the effect of pressure on the wall breaking effect of Ganoderma lucidum spore powder extraction,the extraction pressure increased,resulting in an increase in the pressure difference between the Ganoderma lucidum spore cells inside and outside,which in turn led to rapid cell rupture in a short time,Ganoderma lucidum spore powder The active ingredients of the extract are taken out of the cell with the extractant and diffuse into the solution to achieve the extraction process.For GLT,a standard curve for detecting triterpene content was established by the perchloric acid-ursolic acid-vanillin method,which was used to determine the extraction rate of GLT.Through the single factor experiment,the regularity of the extraction of GLT was summarized by various factors,and the optimal process operating parameters for the extraction of GLT were analyzed by the response surface analysis: pressure 378 MPa,liquid-solid ratio 20 mL/g,holding time 6 min,ethanol volume fraction 75 %,and GLT yield 0.11 %.To establish a kinetic model for the process of ultra-high pressure extraction of GLP.In addition,under the conditions of process operation,the experimental data of the mass concentration of GLP was extracted and compared with the simulation value obtained by the kinetic model.The relevant calculation coefficients were all above 0.95,indicating that the kinetic model of the ultra-high pressure extraction technology was established.It can accurately describe the dynamic process of ultra-high pressure extraction of GLP,and plays a good role in guiding and predicting the application of this process.
