This paper originated from red pigment produced by fungus Paecilomyces lilacinus TD16.Firstly,effects of surfactants on biomass and pigment production were investigated.Then,the extraction and purification process of pigment and pigment structure were studied emphatically.Finally,dyeing and antimicrobial properties of pigment were analyzed.By studying the effects of four different types of surfactants on biomass and pigment production,cationic surfactant CTAB and anionic surfactant SDS were found to have toxic side effects on the growth of organisms and thus reduce significantly the pigment production.Tween-80 and Triton X-100,two kinds of non-ionic surfactants,had little effect on biomass.But Triton X-100 reduced significantly the pigment production.The production of pigment increased with increasing concentration of Tween-80 up to 2.0 g/L.However,a further increase in the Tween-80 concentration led to a marked drop.The maximum production of pigment was achieved at 2.0 g/L,corresponding to significant increases of 108%compared with the control.The methods of extraction and purification of pigment were improved and a high performance liquid chromatography?HPLC?method was established.The best conditions?Kromasil C18,acetonitrile:water=5:3?for HPLC analysis were proposed.Under these conditions,the components of the pigment solution were completely separated.The peak area of retention time at 8.536 min is maximum,which is considered as the main component of pigment.Qualitative analysis suggested that the TD16 pigment was hydroxyanthraquinone compound,and the conclusion was also verified by the spectrums of UV-vis,IR and MS.Moreover,the IR spectrum also indicated that the side chains of TD16 pigment contained amido bond,multiple CH2 and CH3 groups except for the mother nucleus of anthraquinone and phenolic hydroxyl groups.The MS spectrum indicated that the molecular weight of TD16 pigment was 396.01 with the molecular formula of C22H22O6N.To the best of our knowledge,it was reported for the first time.Single factor experiments and response surface methodology were used to optimize the staining technique.Optimum technological conditions were followed as:substantive staining,pH 6,dyeing time 60 min,temperature 52.93?,liquid ratio 1:43.98,pigment concentration 0.44 g/L.Under these conditions,the dyeing rate was 58.44%,which was consistent with the predictive value of the model in response surface,indicating that the model was accurate and reliable.The inhibition effects of dyed textiles on three kinds of bacteria tested were determined by AATCC-100 test Method.The inhibition effect of dyed textiles on gram-positive bacteria was better than gram-negative bacteria.In addition,the inhibition rate of dyed textiles was positively correlated with dyeing rate.
