Rubus suavissimus S.Lee,commonly known as sweet tea,Guangxi sweet tea,has a long history of folk application.It is also used as a tea and sugar substitute product,and used for medicinal purposes.It has the functions of clearing away heat and moistening the lungs,relieving cough,reducing swelling and soothing muscles.The main active substance in sweet tea is rubusoside,which also contains chemical components such as polyphenols,alkaloids and amino acids.Previous studies have shown that rubusoside has a certain effect on intervene in diabetes,but the effect of low concentration is not obvious.In order to effectively develop and utilize the rubusoside of sweet tea,we established a rat model of type II diabetes and explored a suitable dose of rubusoside to alleviate the condition of diabetic rats.On this basis,the extraction process of sweet tea was studied.The extraction method and extraction reagents and dosage,extraction time,temperature and other process parameters were determined,and made a sweet tea lozenge.The following results were obtained:1.A stable diabetic rat model was constructed.Measuring and analyzing the indicators of body weight,blood glucose,serum insulin level,serum triglyceride and total cholesterol.The results showed that one-time tail vein injection of A dose(38mg/kg)and a one-time tail vein injection of C dose(70mg/kg)were successfully building the model.Streptozotocin can successfully construct a diabetic rat model,in which A dose is biased towards the construction of type II diabetes model,and C dose is biased towards constructing type I diabetes model.2.The effect of rubusoside on type ? diabetes was studied.Through the determination and observation of body weight,blood glucose,serum insulin level,serum triglyceride and total cholesterol,islet histopathological staining and other indicators,it is shown that the 100mg/kg dose of rubusoside has significant hypoglycemic effect on diabetic rats,the repair effect of islet tissue is obvious.3.The extraction method of rubusoside was studied.The results showed that theextraction method of rubusoside in sweet tea leaves was better by using the high temperature reflux extraction while 50% ethanol as the extraction solvent.The orthogonal test was used to investigate the temperature,time and solid-liquid ratio,to optimize the reflux extraction process.The results showed that when extraction time is 3 hours,extraction temperature is60? and ratio of material to liquid is 1:8,the extraction rate of rubusoside is high,the stability is good,and the operation is simple.4.The sweet tea extraction was subjected to flavoring research and a sweet tea lozenge was developed.The results of flavor study showed that chitosan and ?-cyclodextrin could be used to treat the sweet tea extract,which could improve the taste.0.7g/L chitosan was used to treat polyphenols in sweet tea extract,the removal effect is better.And use 0.4~0.5g/L?-cyclodextrin improves the taste.The sweet tea extract is made into a sweet tea lozenge by wet granulation method.It is estimated that each 0.5g tablet contains about 0.175 g of rubusoside,and if it is to achieve the hypoglycemic effect equivalent to the hypoglycemic test of the animal,the daily content is take 6~8 slices of sweet tea lozenge.
