As the main force of natural medicine,currently researches of traditional chinese medicine mainly focus on macromolecule substances and few focus on its small molecular substances,leading to some valuable plant resources being ignored.Insomnia is recognized as a multiple health problem that affects modern people and are frequently treated with oral or injection of chemical synthesis drugs.Although these methods have effect,to some extent,there are certain disadvantages.It’s necessary to find natural drug to supplement therapy and traditional chinese medicine is a critical research and application object in the field of sedative and hyponotic.The paper examined the sedative-hypnotic action of inhaled the eupatorium fortunei turcz.essential oil(eeo)and polygala tenuifolia willd.Essential oil(peo).Both of them are from commonly used perennial herb plants and extracted in the laboratory environment.Eupatorium fortunei turcz.Is cultivated in southern china.Polygala tenuifolia willd.Is cultivated in nouthern china and it’s aqueous extracts has been used as a type of sleep-aid medicine in china.The paper first combined the chemical compositions of the essential oil,the animal behavior test,and the nerve function test to explore the possibilities on sedation and hypnosis for inhaling the two natural drugs and increasing the types of plants in rehabilitation garden.The main results are as follows:The chemical constituents analysis of eeo and peo:there were five chemical constituents in the top twenty chemical compounds of eeo,which were sesquiterpenes(39.26%),esters(11.74%),phenols(0.61%),sesquiterpene alcohols(0.61%),and ethers(0.61%).β-sesquiphellandrene are in the first place.Β-caryophyllene are in the second place,linalool are in the seventh place.They all have been reported to have sedative and hypnotic effects on central nervous system.There also were five chemical constituents in the top twenty chemical compounds of peo,which were acids(80.49%),esters(6.42%),monoterpene alcohols(1.04%),sesquiterpenes(1.05%),and aldehydes(0.85%).None of chemical compounds have been reported to have sedative and hypnotic effects.Animal behavioral test:locomotion,pentobarbital sodium-induced sleeping time,and body temperature drop are a classic triad of research methods to determine whether the subjects drugs have sedative effect or not.The results show that eeo and peo have varying degrees of sedative effect.The three concentrations of peo in the study can’t significantly shorten the sleep latency of mice.The results of inhaled eeo(2×10-3g/ml)show that is possibly sedative,reducing the total distance of movement,slowing down speed of movement and prolonging the resting time in mice.At the same time it can also prolong the duration of sleep and decrease the body temperature significantly.The results above are same as the diazepam group mice.Neurochemical experiment:the chronic constriction method was used to make the model to evaluate hypnotic effect of eeo(2×10-3g/ml)in mice.After binding 9 hours per day for 7 days,the sleep-wake rhythm is the most disordered in model and the insomnia is the most serious.Compared with the mice in blank control group,the sleep latency was prolonged by 39%,sleep duration was shortened by 20%in model mice.The normal growth of the mice that were treated with the insomnia model was significantly inhibited compared with the normal group.At the end of the modeling,the locomotion of mice in the control group was significantly higher than that in the normal feeding group,and the locomotion was significantly decreased after the treatment of inhaling eeo.Compared with the insomnia model group,inhaled eeo significantly decreased the level of ne in the striatum,and significantly decreased the metabolic rate of the 5-ht in the hypothalamus and thalamus.The results maybe indicated one of the mechanisms of the sedative-hypnotic efficacy of inhaled eeo.
