The Study Of Tissue Culture Technology And Standardized Production Procedure Of Dendrobium Officinale

This article mainly studied to develop tissue culture technology of Dendrobium.officinale and make standardized production procedures(SOP)of Dendrobium.officinale,the following two aspects is included.(1)By adding different concentration of growth regulator and organic additives on MS basal medium,oberve their effects on Dendrobium seedlings grown from seed to growth.Selected the best suitable ratio of hormone concentration for seed germination,proliferation and differentiation of the original bulb,and rooting.In order to reduce aseptic growth cycle and costs and improve transplanting planting survival rate significantly.(2)Through examining Dendrobium planting base and observing the whole process from the seedling to harvest of Dendrobium,and studing the various stages of Dendrobium(such as transplanting seedlings planting techniques,irrigation and drainage measures,duration of ventilation,humidity,etc.).Initially worked out the standardized production procedure(SOP)of Dendrobium,including field management,disease prevention and pest control,harvesting origin primary processing and other aspects of Dendrobium.Through research,the results are as follows:(1)The most suitable conditions for the original bulb induced is MS + 6-BA0.5mg / L + NAA0.5mg / L + sucrose 30 g / L + agar7.5g / L;The most suitable conditions for the proliferation of PLB is MS + 6-BA0.5mg / L+ NAA0.1mg / L + sucrose 30 g / L + agar 7.5g / L;The most suitable conditions for differentiation is MS + 6-BA0.5mg / L + NAA0.1mg / L + sucrose 30 g / L + agar 7.5g / L;the most suitable conditions for rooting: MS + 6-BA0.5mg / L + NAA1.0mg / L + sucrose30 g / L + agar 7.5g / L(2)Combined with the actual scientific and technological achievements,developed a standardized production procedures guiding significance.
