Agarics blazei is a kind of high temperature fungus,which has very high officinal value,growing mainly in summer and autumn,high demand for temperature,short growing period,and the mycelium autolysis will occur in low temperature storage conditions.In order to preserve Agarics blazei strains better,Agarics blazei strain was used as the test material in this experiment,which was from Microbiology laboratory,College of agriculture,Yanbian University.,to explore the effects of different treatments on carbon and nitrogen utilization ability of Agarics blazei strains,and the influence of several isoenzymes in the mycelium of Agarics blazei belt(esterase isozyme,peroxidase isozyme,protease).After measuring,accounting and analyzing the Agarics blazei growth rate,Polyacrylamide gel electrophoresis test was carried out on the above isozymes,and the enzyme bands and mobility were compared.The results are as follows:1.As the study on Agarics blazei strains' utilizing the source of carbon shows,the strains grow most stably and rapidly with glucose as the source of carbon,followed by sucrose.It reflects glucose and sucrose can be used as the source of carbon in the medium.When studying the Agarics blazei strains' utilizing the source of nitrogen,the most stable and rapid growth of strains occurs in the peptone,followed by soybean peptone.It demonstrates peptone and soybean peptone can be used the source of nitrogen.Agarics blazei strains are kept in the low-temperature conditions,with 20%trehalose solution added as the protection solution,which can improve the Agarics blazei strains utilization of the source of carbon.2.Low temperature imposes a great impact on the activity of mycelial polyphenol oxidases and has an insignificant impact on the activity of?-1,3-glucanase.The activity of mycelial polyphenol oxidase reaches highest when 20%trehalose solution is added as the protection solution.The activity of ?-1,3-glucanase is similar to that of the room-temperature control group.The activity of mycelial polyphenol oxidases treated with sterile water is lower than that without sterile water treatment.It reveals that Agarics blazei strains in the low-temperature sterile water can inactivate the enzyme while the addition of 20%trehalose solution as the protection solution can improve the enzyme activity.3.Low temperature has a considerable impact on the isozyme of Agarics blazei esterase.The longer it is treated in the low temperature,the lower the activity of the esterase isozyme is and the lighter its color is.The addition of the solution has some impact on the acitivty of the isozyme of Agarics blazei esterase.Under the same low-temperature conditions,the acidity of esterase isozyme with solution reaches highest and the activity of esterase isozyme treated with 20%trehalose solution is higher than that of it treated with sterile water.As it suggests,20%trehalose solution can protect mycelium in the low temperature.4.Low temperature has certain impact on the isozyme of Agarics blazei esterase.With the treatment under low temperature gets longer,the color of enzyme bands becomes lighter.The addition of the solution affects the activity of the isozyme of Agarics blazei esterase.In the low-temperature conditions,the activity of strains peroxidase isozymes is the highest.The activity of peroxidase isozymes treated with 20%trehalose solution is higher than that treated with sterile water.It is proved that 20%trehalose solution can protect mycelium in the low temperature.5.Low temperature has certain impact on the Agarics blazei protease isoenzyme.The addition of the solution can impact the activity of mycelial protease isoenzyme.In the low-temperature conditions,the activity of strains ' protease isoenzyme without solution is the highest.The activity of peroxidase isozymes treated with 20%trehalose solution is higher than that treated with sterile water.The new low-temperature-inducible protein appears in the low temperature.The relationship of protein nature and low temperature needs further studies.
