In order to study association of the A-FABP,POMC and UCP3 gene variations with some meat production traits in cattle,which can provide a theoretical basis for improved varieties selection of cattle,in this study,the polymorphism of 3 candidate genes including A-FABP,POMC and UCP3 gene in 318 cattles including Angus Cattle,Hereford Cattle,Jinnan Cattle,Limousin Cattle,Luxi Cattle,Qinchuan Cattle,Simmental Cattle and Charolais Cattle were analyzed by PCR-SSCP or PCR-RFLP,and association analysis of their polymorphisms with meat production traits were analyzed by developing an association model.The research result shows as follows:1.The genetic polymorphism of A-FABP,POMC and UCP3 gene.Nine mutations were detected in this three genes which including A2989G,A3533T and T4014C in A-FABP gene,T5141C,G5785C and C6621G in POMC gene,G4921A,C4524T and A5191G in UCP3 gene.All of the above.SNPs were divided into three genotypes,respectively.2.Association analysis of A-FABP gene with meat production traits in cattle Polymorphism of A2989G mutation in A-FABP gene is significantly associated with marbling score and is highly significant associated with loin muscle area.Both marbling score and loin muscle area of AG and GG genotype individuals were extremely significantly higher than that of AA genotype(P<0.01),while there was no significant difference between AG and GG genotype individuals(P>0.05).Individuals with the G allele were extremely significantly higher(P<0.01)than the others in marbling score and loin muscle area.Polymorphism of A3533T mutation in A-FABP gene is significantly associated with carcass weight and waist muscle thickness,which includes,carcass weight of AA genotype individuals were extremely significantly higher than that of AT genotype(P<0.01),while waist muscle thickness of genotype AT were significantly higher than that individuals with genotype AA(P<0.05).Different genotypes of this locus have different effect on on the two traits.Polymorphism of T4014C mutation in A-FABP gene is significantly associated with meat tenderness,waist muscle thickness and carcass length,which includes,individuals with CC or TC genotype were extremely significantly higher(P<0.01)or significantly higher(P<0.05)than that of TC genotype in meat tenderness,respectively,individuals with CC genotype were extremely significantly higher(P<0.01)or significantly higher(P<0.05)than that of TT or TC genotype in waist muscle thickness,respectively,while carcass length of CC and TT genotype individuals were significantly longer than that of TC genotype(P<0.05),CC genotype is improving carcass length,meat tenderness and waist muscle thickness.3.Association analysis of POMC gene with meat production traits in cattle Polymorphism of T5141C mutation in POMC gene is significantly associated with live weight and meat tenderness,which includes,live weight of CC genotype individuals were significantly longer than that of TC genotype(P<0.01),individuals with CC genotype were extremely significantly higher(P<0.01)or significantly higher(P<0.05)than that of TC or TT genotype in meat tenderness,respectively.Individuals with CC genotype were significantly better than others in meat tenderness and live weight.Polymorphism of C6621G mutation in POMC gene is significantly associated with loin muscle area and thigh meat thickness,which includes,individuals with CC or CG genotype were significantly higher(P<0.05)or extremely significantly higher(P<0.01)than that of GG genotype in loin muscle area,respectively,thigh meat thickness of CG genotype individuals were significantly higher than that of GG genotype(P<0.05).Individuals with CG genotype were significantly better than others in loin muscle area and thigh meat thickness.4.Association analysis of UCP3 gene with meat production traits in cattle Polymorphism of G4912A mutation in UCP3 gene is significantly associated with backfat thickness,which includes,individuals with GG genotype were significantly higher than that of GA genotype in backfat thickness(P<0.05).Polymorphism of A5191G mutation in UCP3 gene is significantly associated with meat percent,loin muscle area and average daily gain,which includes,individuals with AG and AA genotype were both extremely significantly higher than that of GG genotype in meat percent(P<0.01),individuals with AG genotype were significantly higher than that of GG genotype in loin muscle area(P<0.05),individuals with AG or AA genotype were extremely significantly higher(P<0.01)or significantly higher(P<0.05)than that of GG genotype in average daily gain,respectively.Individuals with the A allele were significantly higher than the others in meat percent,loin muscle area and average daily gain.Conclusion was drawn that the A2989G and T4014C mutation in A-FABP gene,the T5141C and C6621G mutation in POMC gene and the G4912A and A5191G mutation in UCP3 gene can be used as molecular marker of meat production of beef cattle.
