Hypsizigus marmorens is one of rare and gracious fungi with low-fat,low-calorie and high-protein.However,due to long growth cycle,low yield and less resistance ability,in addition,the increase of material and transportation costs with Hypsizigus marmorens.The economic benefit of enterprises engaged in Hypsizigus marmorens production get bigger impact.In this study,according to the parent strains Hypsizigus marmorens and the fusion strains Minzhen two with mycelia,fruiting body of material,using difference proteomics method analyze the different protein spots of mycelia and fruiting body from the fusant and the parent,comparing the proteins of biological characteristics.While taking advantage of abundant and cheap agriculture wastes including loquat kernel,grain stillage,tea-leaf from local company were used to replace cornflour,sawdust and wheat bran in the previous culture medium,The optimal medium components were obtained.The results are as follows:According to the parent strains Hypsizigus marmorens and the fusion strains Minzhen two with mycelia,fruiting body,46 protein spots were haved by using PDQuest8.01 software analyze the Hypsizigus marmorens differential proteins in parent and fusant,using MALDI-TOF/TOF-MS/MS mass spectrometry get 38 MS signals,search the NCBInr database successfully identified 28 proteins,five similar functional proteins,five proteins of unknown function.These involved in translation elongation,hydrolyzed protein folding,stress response,glycolysis,signal transduction,energy metabolism.Serine proteases and ketol-acid reductoisomerase play a role of signal transduction in Hypsizigus marmorens growth.Heat shock protein HSP 70 and NAD+ dependent formate dehydrogenase involved in stress responses of Hypsizigus marmorens growth,the expression of these two proteins in fusant is higher than the parent,shows the fusant environmental adaptability,resistance is stronger than the parent,Enolase,phosphopyruvate hydratase,voltage-dependenion-selective channel,S-adenosyl-L-homocysteine hydrolase,fructose-bisphosphate aldolase,control the Hypiszigus marmoreus growth through energy metabolism,substance metabolism in the glycolytic pathway.These types of protein expression is higher in the fusant,shows the more active energy metabolism and substance metabolism regulation in fusant.Cyclophilin protein involved in protein folding,the proteasome-domain-containing protein and disulfide isomerase as molecular chaperones decomposited incorrect folded proteins,increasing the cell stability and reducing abnormal growth.Catalase,cysteine peroxiredoxin clear Hypiszigus marmoreus's peroxide during growth,delay cell senescence.The expression of these proteases in the fusant is more than parent,shows the more stable,the batter production performance in fusants growth.Through a comprehensive analysis of Minzhen two different combinations between the mycelia covered with bags of time,the number of effective duo,cap diameter,stipe length,yield and biological efficiency of the combination 1 the least time of mycelia covered with bags,the moderat number of effective duo,the highest of cap diameter,stipe length,yield and biological efficiency.Therefore,the most suitable for cultivation formula for Minzhen two is combination I:a composition,54%cotton shells,3%loquat core,10%tea-leaf,15%grain stillage,15%corn cobs,2%lime,calcium carbonate 1%and moisture 65%.
