| In this paper,phylogenetic relationships and biological properties(morphological characteristics,rhizosphere microbial diversity,nutritional value,resistance and et.)of 5 cultivated JUNCAO varieties(JC01,JC02,JC03,JC04,JC05)screened from the wild do a preliminary study.Determine its taxonomic status and basic biological properties.The main results are as follows:(I)Phylogenetic relationships among 5 cultivated JUNCAO varieties screened from the wild are studied by ITS1-5.8s-ITS2 rDNA sequences.Results show that the length of cloning,sequences are 634-642bp,and the genetic relationship amount 5 JUNCAO strains were analyzed by cluster.Combined with the results of the morphological identification,we preliminarily confirm that JC01,JC02,JC03 and JC04 are Arundo and JC05 is Pennisetum.They are quite different from existing varieties and therefore may be new cultivated JUNCAO varieties.(2)Determine the number of rhizosphere soil bacteria,fungi,actinomycetes from 5 cultivated JUNCAO varieties screened from the wild by plate culture method.Results show that:The number of rhizospheric microorganism from 5 cultivated JUNCAO varieties screened from the wild planting six months were significantly higher than the control groups,and all of them reached a very significant level.(3)Compare the contents of moisture,ash,crude polysaccharides,crude protein,crude fat,crude fiber from 5 cultivated JUNCAO varieties screened from the wild.Results show that:In the moisture content,JC05>JC02>JC01>JC04>JC03.The maximum reached 80.77%is JC05.The minimum reached 72.13%is JC03.In the content of ash,JC05>JC03>JC02>JC01>JC04.The maximum reached 9.41%is JC05.The minimum reached 7.93%is JC04.In the content of crude polysaccharide,JC01>JC02>JC04>JC05>JC03.The maximum reached 5.49%is JC01.The minimum reacheed 4.98%is JC03.In the content of crude protein,JC01>JC04>JC03>JC05>JC02.The maximum reached 16.13%is JC01.The minimum reacheed 10.85%is JC02.In the content of crude fat,JC02>JC04>JC01>JC03>JC05.The maximum reached 6.84%is JC01.The minimum reacheed 2.38%is JC03.In the content of crude fiber,JC01>JC04>JC03>JC05>JC02.The maximum reached 33.37%is JC01.The minimum reacheed 32.01%is JC02.(4)Determine the leaf net photosynthetic rate(Pn)of 5 cultivated JUNCAO varieties screened from the wild in the artificial simulated stress.Results show that:In high temperature stress,JC05>JC04>JC03>JC02>JC01.The maximum reached 64.56μmol·m-2·0-1 is JC05.The minimum reached 4.16 μmol·m-2·s-1 is JC01.In low temperature stress,JC02>JC01>JC05>JC04>JC03.The maximum reached 13.5 μmol·m-2·s-1 is JC02.The minimum reached 8.42 μmol·m-2·s-1 is JC03.In drought stress,JC05>JC01>JC04>JC03>JC02.The maximum reached 26.72 μmol·m-2·s-1 is JC05.The minimum reached 9.16μmol·m-2·s-1 is JC02. |
