In recent years,the economic and medicinal value of Bletilla striata is getting higher and higher.At present,Bletilla striata seedlings mainly use tissue culture,there are many problems such as medium pollution,long culture time and so on.How to improve the number of seedlings and the quality of direct seeding is an urgent problem to be solved.At present,a large number of compound fertilizers are mainly used,which have high cost and influence on the quality of Bletilla striata,while biogas manure is rich in nutrients needed in plant culture,which has the characteristics of slow-acting fertilizer and low cost.Therefore,biogas manure is widely used in agricultural production.If the biogas manure can be successfully applied to direct seeding and planting with Bletilla striata,it can not only improve the utilization rate of biogas manure,reduce the environmental pollution,but also save the cost of breeding and planting of Bletilla striata.Therefore,this paper studies the application of biogas manure in the direct seeding of Bletilla striata,the planting of Bletilla striata and the experiment of Bletilla striata planting in order to provide theoretical guidance for the standardization and industrial cultivation of Bletilla striata.The main results are as follows:(1)In the direct seeding experiment of Bletilla striata,the effects of substrate,hormone,fertilizer(biogas fertilizer?water-soluble fertilizer)on germination and growth of Bletilla striata were investigated.The results showed that a thin layer of sand was sprayed on the surface of the prepared substrate,and a small amount of hormone 6-BA2.0mg·L~-11 was sprayed.The peat with 65%moisture content was used as the substrate,and the temperature was controlled at 30?for 16 h.The seed comprehensive growth condition reached the best;The effect of biogas slurry on the germination of Bletilla striata was the most significant,and the biogas residue had the best effect on the growth of Bletilla striata at the later stage of germination,and the germination rate of Bletilla striata could reach 95%when the concentration of biogas liquor was 40%(diluted with the original biogas liquid).The germination rate of Bletilla striata reached 87%when the amount of biogas dregs was about5%(in percentage of substrate volume),and the seedlings grew strong,dark green and pseudobulbous at the later stage.(2)In the pot experiment of Bletilla striata,the effects of culture medium and fertilizer on the growth of A.splendens seedlings with seedling age of one year were investigated.The results showed that the best culture medium was soil cultivation,the maximum leaf length and width,and plant height were the best.The best fertilizer for Bletilla striata was water-soluble fertilizer.Leaf characters of aboveground part:from May to June,0.6%(percentage of substrate volume)was the best,and the leaf length and width increased by 9.6 cm and 0.63 cm,respectively.From June to August,the optimal growth rate was 1%,with a growth rate of9.6cm in leaf length and 0.4cm in width.From may to August,the optimum plant height was0.6%with an increase of 6.33cm.The optimum stem diameter was 1%and the growth was0.27cm.For the underground part,the optimal dosage was 1%,with the tuber length(5.35±0.65a)cm and width(2.05±0.05a)cm.The highest content of Bletilla striata polysaccharide in biogas slurry was 3.15%.The correlation analysis shows that the diameter of Bletilla striata is significantly correlated with the size of tuber.(3)In the study of Bletilla striata application,the effects of three fertilizers(biogas slurry,biogas residue and water-soluble fertilizer)on the growth of Bletilla striata,which were three years old,were tested in greenhouse and three-dimensional cultivation.The results showed that470g/m~3 was the best fertilizer and dosage of Bletilla striata.The effects of different layers on the growth of Bletilla striata were investigated in three-dimensional culture.The results showed that the upper layer of the shelf was the best.From May to June,the growth of large leaf length was 3.83 cm,the width of large leaf was 0.6 cm,from June to August,the growth of the big leaf length was 3.5cm and the width was 0.5cm.From may to August,the plant height increased by 6.25cm.Stem diameter,growth up to 0.07cm;For the underground part,the comprehensive score was 95,in which the tuber length was(3.33±0.34a)cm.Finally,it was found that the overall agronomic characters of Bletilla striata under stereoscopic cultivation were lower than that of pot and field cultivation,but the yield rate was significantly higher than that of pot and field planting,so the three-dimensional cultivation frame was improved.At the same time,it was concluded that Bletilla striata seedlings were three-dimensional cultivation and water-soluble fertilizer was applied.It is the best way to plant Bletilla striata by applying water soluble fertilizer(ornamental)/biogas manure(medicinal)and applying biogas dregs to the seedling age of three years in greenhouse pot culture/field culture at the later stage.The results of this study can provide theoretical guidance for direct seeding and large-scale planting of Bletilla striata.
