Pleurotus ostreatus is the biggest cultivated size edible fungus in China.Composted cultivation and sterilized cultivation are the most common used cultivation styles of Pleurotus ostreatus production.The quality of composted substrates is a key factor to success rate of cultivation bags and production.Composting time also shows effect on the sterilized cultivation.Corncob and cotton seed hull were used as the main substrates in this study,the physiochemical property changing patterns of composted corncob and cotton seed hull,effect of composting time on the sterilized cultivation of Pleurotus ostreatus were studied,effect of different formulations on composting cultivation of Pleurotus ostreatus,The main results are as follows:1.Composting time of corncob on Pleurotus ostreatus cultivation experiment results showed: In the composting process of corncob,substrate color turned from golden to puce,along with ammonia flavour disappearing,particle size diminishing and viscosity increasing,temperature increased first and decreased later,p H value and water content gradually decreased,The number of bacterias,fungis and actinomycetes increased first and decreased later,cellulose content and hemicellulose content increased first and decreased later,Stacking,the percent content of cellulose and hemicellulose was 34.8% and 36.7% respectively,Before bagging,the content decreased to 24.9% and 21.6% respectively.And lignin content increased gradually,Increased from 18.4% of stacking to 30.2% of before bagging;total carbon content decreased gradually,Decreased from 44.6% of stacking to 38.7% of before bagging and total nitrogen content decreased first and increased later and decreased again,Among them,the content reached the highest during the third turning,was 1.12%.Meanwhile,composting time of corncob experiment results showed that the best composting time of corncob for subsequent sterilized cultivation was 6 days,which resulted in a highest biological efficiency,was 89.74%.In the whole growth and development process of Pleurotus ostreatus,cellulase and semicellulase activities increased first and decreased later,and reached their peak at primordia stage of the first flush,while laccase activities showed a decreased tendency.In the same growth and development stage,the activities of those three enzymes decreased along withwith the elongation of composting time.2.Composting time of cotton seed hull on Pleurotus ostreatus cultivation experiment results showed: In the composting process of cotton seed hull,substrate color turned from bronzing to black brown,along with ammonia flavour disappearing,substrate size diminishing and viscosity increasing,temperature increased and down later,and decreased faster than that in corncob composting process.The composting time was shorter than that in corncob composting process.p H and water content gradually decreased.The number of bacterias,fungis and actinomycetes increased first and decreased later.Cellulose content and semicellulose content gradually decreased,during stacking,The percent content of cellulose and hemicellulose was33.6% and 27.8% respectively,Before bagging,The content decreased to 23.9% and14.9% respectively.And Lignin content gradually increased,Increased from 20.2% of stacking to 26.28% of before bagging;Total carbon content gradually decreased and total nitrogen content gradually decreased through a fluctuating way,Stacking,the percent content of total carbon and total nitrogen was 47.1% and 1.27% respectively,Before bagging,the content decreased to 43.9% and 1.13% respectively.Meanwhile,composting time of cotton seed hull experiment results showed that the best composting time was 5 days,which resulted in a highest biological efficiency,was 93.75%.In the whole growth and developmental process of Pleurotus ostreatus,cellulase and semicellulase activities increased first and decreased later,and semicellulase activities reached its peak when the mycelia spread full of the cultivation bag.Cellulase activities reached its peak at the primordia stage of the first flush,while laccase activities showed a gradual decreased tendency.At the same growth and development stages,the activities of those three enzymes decreased along with the elongation of composting time.3.The results of formulation experiments indicated that formulation 2 consisting of cotton seed hull 96.7%,urea 0.3%,lime 3%,water content 65%,gave the highest average yield and biological efficiency,were 1.23 kg and 100.36% respectively.but to consider economic benefit,formulation 1,which consisted of corncob 90.5%,urea1.5%,lime 4%,calcium magnesium phosphate fertilizer 4%,water content 65%,gave the highest economic benefit,and the average profit per bag is 2.5 yuan. |
