Impatiens is one of the largest genera,containing over 1000 species.The plants of the genus are annual or perennial herbs.Their stems often succulent and succulent.The morphological variation of Impatiens is extremely complex,their geographical distribution show distinct regional characteristics and unique phenomenon,These factors made Impatiens becoming a very difficult group in plant taxonomy.Researcher of this genus has put forward some classification systems or auxiliary unit under the genus(group or subgenus),but has not been accepted by most scholars.Up to now,there is not a recognized classification criteria and classification system.Impatiens in China have been found more than 270 kinds,mainly of them distributed in the southwest of China.The vast majority of Impatiens are endemic to the narrow domain in China.Along with the further investigation of the field,many new species have been reported.This research mainly relies on a basic work of the National Science and Technology Department: a comprehensive study on the biological diversity in Wuling mountain area.Taking the investigation of Impatiens as my main task in field.The works included collecting photos and other data samples.To stud on the molecular phylogeny relationships for reveal the Impatiens development.In this paper,the author uses the method of FIASCO(Fast Isolation by mark ers of Sequence Containing Repeats)to build the enriched microsatellite library in I.Fenghwaiana,following to get sequence of containing SSR(Simple Seq uence Repeat).The results of the study are as follows:1.In the early stage,the author put the focus on the field investigation in the western hubei,hunan,tongren city and other places of the Wuling mountain area.In the process,we found 21 species of Impatiens and nine new record species of Hubei province: I.siculifer var.porphyrea,I.fenghwaiana,I.pinfanensis,I.bodinieri,I.guizhouensis,I.taishunensis,I.paradoxa,I.alpicola,I.Conaensis.2.Based on ribosomal ITS sequence and two gene sequence of chloroplast,the author used the ways of sequence characteristics analysis,genetic distance analysis,phylogenetic tree research.In the study phylogenetic,the auther selected Hydrocera triflora,tetramerista sp.as the outgroup to established tree by the ways of neighbor joining(NJ tree),maximum parsimony(MP tree)and Mrbayes software(Bayes tree).The analysis indicated that Hydrocera triflora is lower than Impatiens in the process of evolution,and they belong to the sister group.The Impatiens pritzelii,which have 4 carpels,is quite near Hydrocera triflora in genetic distance.In other words,the results support that 5 carpels of ovary were evolved from 4 carpels in the Impatiens.Meanwhile,the ITS sequences are relatively conservative,then it makes little sense to study system evolution in Impatiens.For another,the psbA-trnH gene sequence were applied to the taxonomic researches of Impatiens for the first time.3.In this study,13 polymorphic microsatellite markers were developed from I.Fenghwaiana genomic libraries using the fast isolation by amplified fragment length polymorphism of sequences containing repeats method.The high cross-species transferability of these markers was evaluated in five other Impatiens species: I.Balsamina,I.noli-tangere,I.Blepharosepala,I.Oxyanthera,I.Davidi.Among them,there are 3 markers showed polymorphism in five other Impatiens species.This is the first plants of the genus for the development of molecular markers,develop these polymorphic SSR markers to the ecology and population genetics of the genus study provide important basis.This is the first time for molecular markers of Impatiens,and these microsatellite markers will provide powerful basis for population genetic and ecological studies of I.Fenghwaiana and its related species in the future.
