Corylus hetercphylla x Corylus avellana has beautiful branches,strong drought resistance and cold resistance.Its fruit has high yield,high nutritional value.So it was excellent tree species both have ornamental value and economic value.The annual hardwood and softwood of Corylus heterophylla x Corylus avellana were selected as cuttings,the effects of different hormone concentrations and media were applied on cuttings in container on rooting were investigated.The results were as follows:1.The rooting rate of hardwood cuttings was improved by hormones.The results showed that ABT 1 with 100 mg·L-1 for 30min was the best in all the treatments,the rooting percentage was 77.22%,average primary lateral root number of cuttings was 10.33,average root length was 8.25 cm,the thickest root diameter was 1.91 mm,compared with the control significantly increased by 22.19 times,2.87 times,2.99 times,3.55 times at 0.01 level,average secondary lateral root number of cuttings was 51.00;the rooting effect of IBA alone or compound hormone treatment was poor,the rooting rate was slightly higher at 20%~25.00%when treated with 400 mg·L-1 IBA and 100 mg·L-1 IBA +25 mg·L-1 ABT 1.2.The different ratio of media had significant effects on rooting for hardwood cuttings.It is best to use the No.3 media(V sand:V peat:V perlite =1:3:3).The rooting percentage was 71.11%,average primary lateral root number of cuttings was 13.33,average root length was 7.29 cm,the thickest root diameter was 2.10 mm,average secondary lateral root number of cuttings was 58.45.The rooting rate of cuttings in other media was 20.00%~58.89%.3.The effect of different hormone treatments on rooting of softwood was significant.IBA quick dipping treatment significantly improved the rooting effect of cuttings.IBA with 1000 mg·L-1 for 30 second was the best in all the treatments,the rooting percentage was 75.00%,average primary lateral root number of cuttings was 17.67,average root length was 4.56 cm,the thickest root diameter was 1.86 mm.The rooting effect of hormone soaking treatments were poor,the rooting rate was slightly higher at 32.00%~34.67%when treated with 400 mg·L-1 IBA and 200 mg·L-1 IBA +25 mg·L-1 ABT 1 for 3 hour.4.The content of soluble sugar,starch and soluble protein decreased gradually,and the activity of POD and IAAO increased gradually from 0 to 15 days of hardwood.On the 15 th day:compared with the control the soluble sugar content of the cuttings treated with 100 mg·L-1 ABT 1 was significantly reduced by 16.81%at 0.01 level,soluble protein content was significantly reduced by 13.35%at 0.05 level,POD activity was significantly increased by 42.71 times at 0.01 level.The content of soluble sugar in the cuttings decreased continuously,the content of starch and soluble protein increased gradually,the activity of POD increased and the activity of IAAO decreased gradually from 15 to 30 days.On the 30th day:compared with the control the starch content of the cuttings treated with 100 mg·L-1 ABT 1 was significantly increased by 8.91%at 0.05 level,POD activity was significantly increased by 88.92%at 0.01 level,IAAO activity was significantly increased by 2.96%at 0.05 level.The content of soluble sugar,starch and soluble protein in the cuttings decreased gradually,the activity of POD increased and the activity of IAAO decreased gradually from 30 to 45 days.On the 45th day:compared with the control the soluble sugar and starch content of the cuttings treated with 100 mg·L-1 ABT 1 was significantly increased by 25.01%and 33.76%at 0.01 level,POD activity was significantly increased by 1.64 times at 0.01 level.The content of soluble sugar,starch and soluble protein in the cuttings increased gradually,the activity of POD and IAAO increased gradually from 45 to 60 days.On the 60th day:compared with the control the starch content of the cuttings treated with 100 mg·L-1 ABT 1 was significantly decreased by 11.99%at 0.01 level,POD activity was significantly increased by 49.77%at 0.01 level,IAAO activity was significantly decreased by 3.83%at 0.05 level.The effect of the nutrient and enzyme activity in cuttings was small in different media,and the trend was consistent with the hormone test.It can be see that appropriate hormones soaked hardwood cuttings could speed up the consumption of nutrient and the rise of POD and IAAO activity in initial cutting,provided adequate nutrition and energy for callus,and promoted the formation of the callus.At the same time,a lasting nutrition and energy was provided by appropriate hormone treatment for the growth of adventitious roots,so that the consumption of nutrients and accumulation of cuttings gradually gentle,and the POD and IAAO activity decreased,IAA was accumulated,so it was conducive to the formation of adventitious roots;In addition,the appropriate hormone treatment delayed the accumulation of nutrients and the activity of POD and IAAO in the phloem of the cuttings,which was beneficial to the growth of adventitious roots.5.The content of soluble sugar increased gradually,the content of starch and soluble protein decreased gradually,and the activity of POD and IAAO increased gradually from 0 to 15 days of softwood.On the 15 th day:compared with the control the soluble sugar content of the cuttings treated with 1000 mg·L-1 IB A was significantly reduced by 3.81%at 0.05 level,POD activity was significantly increased by 25.33 times at 0.01 level.The content of soluble sugar and soluble protein in the cuttings decreased gradually,the content of starch increased gradually,the activity of POD increased and the activity of IAAO decreased gradually from 15 to 30 days.On the 30th day:compared with the control the soluble content of the cuttings treated with 1000 mg-L-1 IBA was significantly increased by 7.39%at 0.01 level,POD activity was significantly increased by 26.22%at 0.01 level.The content of soluble sugar,starch and soluble protein in the cuttings increased gradually,the activity of POD increased continuously and the activity of IAAO decreased gradually from 30 to 45 days.But compared with the control the soluble sugar and starch content of the cuttings treated with 1000 mg·L-1 IBA was significantly decreased by 22.64%and 31.68%at 0.01 level on the 45th day,the content of the soluble protein was significantly increased by 6.72%at 0.05 level,POD activity was significantly increased by 22.91%at 0.05 level.It can be see that softwood cuttings were soaked by appropriate hormones could speed up the consumption of soluble protein and the rise of POD and IAAO activity in initial cutting.Soluble protein provided a material basis for the formation of root primordia and promoted the formation of callus.At the same time,a lasting nutrition and energy was provided by appropriate hormone treatment for the growth of adventitious roots,so that the consumption of nutrients and accumulation of cuttings gradually gentle.And softwood cuttings were soaked by appropriate hormones could effectively adjust POD activity in cuttings moderate,IAAO activity was reduced,so that the auxiliary factor to promote adventitious root growth was guaranteed,and IAA was accumulated,thus promoted the formation of adventitious roots.Therefore,hardwood cuttings were soaked by 100 mg·L-1 ABT 1 for 3 h,and cutted into the media of V-sand:V-peat:V-perlite = 1:3:3,the rooting was better.Softwood cuttings were quick dipped by 1000 mg·L-1 IBA for 30 s,could achieve better rooting,but also greatly shortened the processing time.
