Prunus Cistena is sheepberry or dungarunga belongs to plum of rosebush,with high ornament al value.Its tree form is compact and new leaves are purplish red.The bark of current branch is red.In order to provide scientific guidance to the production practice of Prunus Cistena,we studie d the effects of different hormone types,concentrations,different medium and their physicochemica 1 properties on the rooting and rooting mechanism in this paper.The main experimental results we re as follows:1.Hormone was treated separately or mixedly,can improve the rooting effect significantly of Prunus Cistena softwood cuttings.Results indicated that the most suitable treatment of low concent ration NAA and ABT 1 soaked separately was 100 mg/L ABT 1 with the rooting rate 85.69%,th e rootnumber 11.17,the average root length 4.82 cm,the longest root length 10.50 cm and the ro ot effect index 1.79 after the cuttings were soaked for three hours,which were increased 10.17,2.04,2.49,1.28 and 4.97 times than that of the control significantly(P<0.01).The most suitable tre atment of low concentration NAA and ABT 1 soaked constant volume mixedly was 50 mg/L NA A+100 mg/L ABT 1 with the rooting rate 90.49%,the root number 16.33,the average root length 4.77 cm,the longest root length 10.87 cm and the root effect index 2.20 after the cuttings were soaked for three hours,which were increased 11.35,3.90,1.06,1.55 and 12.75 times than that of the control significantly(P<0.01).2.The increase of substrate bulk density was beneficial to improve rooting effect of Prunus Cistena hardwood cuttings.Sand,peat,perlite by 3:1:3(V/V)was the suitable media of hardwood cutting,the rooting rate,the root number,the average root length and the longest root length resp ectively to 79.40%,14.45,3.52 cm and 8.28 cm,which were increased 3.40,0.64,0.11,0.29 time s than that of the sand,peat,perlite by 1:1:3(V/V)significantly(P<0.01),the root effect index wa s 1.70,which was increased 5.07 times than that of the sand,peat,perlite by 1:1:3(V/V)significa ntly(P<0.05).Correlation analysis revealed that the greater the density and pH of cutting medium,the higher the rooting rate,the more the root number.But the bigger the EC of cutting medium,the lower the rooting rate,the shorter the average root length and the longest root length.3.According to the rooting site of adventitious roots of cuttings,it can be determined most P runus Cistena cuttings belong to bark and callus rooting of mixed type,had both bark rooting typ e and callus rooting type.The rooting process of Prunus Cistena can be divided into 3 stages,(1)Callus induction and occurrence period;(2)Induction and development of adventitious root primord ia;(3)Adventitious root formation and elongation.4.At the early stage of cuttage.Increasing substrate bulk density was conducive to starch acc umulation,rising of POD activity,reducing of IAAO activity,and thus promoted rooting of Prunus Cistena hardwood cuttings.Starch contents of cuttings in sand,peat,perlite by 1:2:2(V/V)and san d,peat,perlite by 2:3:1(V/V)compared with sand,peat,perlite by 1:3:3(V/V)increased significantl y by 26.64%and 13.66%respectively.The POD activity of cuttings increased significantly by 34.73%and 34.51%respectively.The IAAO activity of cuttings reduced significantly by 10.88%and 12.69%respectively.5.At the early stage of cuttage.Hormone treated separately or mixedly was conducive to acc umulation of soluble protein and soluble sugar and rising of POD activity,and thus promoted rooti ng of Prunus istena softwood cuttings.The soluble protein contents of cuttings treated by 50 mg/L NAA,100 mg/L ABT 1 and 50 mg/L NAA+100 mg/L ABT 1 respectively,compared with con trol increased significantly by 30.80%?1.45%and 25.85%respectively.Soluble sugar contents of c utting increased significantly by 14.50%?13.90%and 10.93%.The POD activity of cuttings increa sed significantly by 27.27%?7.48%and 28.08%.In summary.when Prunus Cistena hardwood cuttage,50 mg/L NAA+100 mg/L ABT 1 was th e most suitable hormone,the most suitable medium was sand,peat,perlite by 3:1:3(V.V)and thec uttings should be soaked for an hour.When Prunus Cistena softwood cuttage,50 mg/L NAA+100 mg/L ABT 1 was the most suitable hormone,the most suitable medium was sand,peat,perlite by 3:1:3(V/V)and thecuttings should be soaked for three hours.The appropriate concentration of AB T 1 and NAA mixed treatment can accelerate the hydrolysis of starch in cuttings can be directlyus ed as a respiratory substrate soluble sugar to provide sufficient nutrients to promote rooting of cutting.
