| The digestive system and immune system of weaning Rex Rabbit are weak,so it can easily occur diarrhea and affect the benefits of farms.Therefore,it is very important to control the weaning stress of Rex Rabbit.The main method of prevention and treatment of Rex Rabbit diarrhea is the use of antibiotics,but the use of antibiotics will produce drug resistance and drug residues and other issues.With the emphasis on food safety,the advantages of traditional Chinese medicine(TCM)gradually highlighted,especially TCM's natural,low toxicity and versatility and other characteristics make it more and more people like it.The purpose of this experiment is to design 3 kind of Sijunzi flavored compounds,then choose the most effective compound by animal test.Then make it into granules by modern pharmacy technology and preliminary evaluate its safety.Finally,feed the weaning Rex Rabbits with granules and observe the effect of Sijunzi flavored granules.The following is theresult:?Screening of Sijunzi flavored compoundIn this experiment,three Sijunzi flavored compounds were fed with weaning Rex Rabbits.The result of the traditional Chinese medicine compound screening test showed that the diarrhea frequency of test group ? was very significantly(P<0.01)lower than control group.The average daily dietary intake(ADFI)and average daily gain(ADG)of test group ? was very significantly(P<0.01)higher than control group.The feed conversion rate(FCR)and serum alanine aminotransferase(ALT)activity of test group ? was significantly(P<0.05)lower than control group.Therefore,the basal diet supplemented with 1%Sijunzi flavored compound ?(Codonopsis,Hawthorn,Atractylodes,Astragalus,Licorice,Bupleurum,Poria,Cudzu)can better reduce the diarrhea frequency of weaning Rex Rabbits,improve their growth performance and could protect their liver function.So the Sijunzi flavored compound ? was choosed to further testing.?Study on extraction technology of Sijunzi flavored compoundIn this experiment,the boiling method was used to extract the active constituents of the Sijunzi flavored Decoction.The extraction yield of Sijunzi flavored decoction was taken as an evaluation index.The results of single factor selected 4 factors to do orthogonal test.The orthogonal results showed that the effect of the boiling time and boiling times on the extraction yield were obvious.The effect of soaking time and adding water times were smaller.The best extraction process of Sijunzi flavored decoction is soaking 2h,adding 8 times water,boiling 3h and boiling 3 times.?The preparation of Sijunzi flavored granule and preliminary evaluation of it's safety.In this experiment,the preparation process of Sijunzi flavored granule was optimized by comprehensive score of forming rate,melting time,hygroscopicity and the bulk density.The result of single factor selected 3 factor including filler selection,the amount of filler and wetting agent(ethanol)concentration.Then optimized thepreparation process of granules by 3 factor and 3 levels orthogonal test.The results showed that the effect of filler selection and the amount of filler on the comprehensive score of granules were obvious,and the effect of the concentration of wetting agent(alcohol)on the comprehensive score of granules was smaller.Finally select soluble starch,filler and exreact powder(1.5:1),60%alcohol.The Sijunzi flavored granule's maximal tolerance dose of the mice was 30.47g/kg(raw drug content).Health effects of Sijunzi flavored granule on weaning Rex Rabbits In this study,we added Sijunzi flavored granule to the basal diet of weaning Rex rabbits to observe it's health effect.The results showed that the diarrhea frequency of 3 test groups were very significantly lower(P<0.01)than control group,the diarrhea frequency of high dose group was lowest and the mortality rate and FCR was significantly lower(P<0.05)than control group.In addition,the basic diet added 2.0%of Sijunzi flavored granule could significantly(P<0.05)decrease weaning Rex Rabbits'serum TC level and ALP activity.The serum ALT and AST activity of high dose group was very significantly(P<0.01)lower than control group.The serum IgG and IgA level of high dose group was significantly higher(P<0.05)than control group.The serum GLU,IgM level and jejunum lipase activity of high dose group were very significantly higher(P<0.01)than control group.Therefore,the basic diet added with 0.5%,1.0%,2.0%of Sijunzi flavored granule could reduce the diarrhea frequency of weaning Rex rabbits and improve their growth performance,promote their absorption of sugars,fats,proteins and other nutrients to a certain extent,protect their liver function and improve their immune function.And the basic diet added with 2.0%of Sijunzi flavored granule could improve weaning Rex Rabbits' health and growth performance better after 30d feeding.Conclusion:(1)The experiment successfully selected the effective Sijunzi flavored compound.It can better reduce the diarrhea frequency of weaning Rex rabbits,improve their growth performance and protect their liver function.(2)The best extraction methods were as follow:add 8 times water,soak for 2h,cook for 3h,cook 3 times.(3)The optimum preparation process of Sijunzi flavored granule were as follow:Soluble starch as the filler,the amount of filler use was filler:extract=1.5:1,wetting agent was 60%ethanol.The results of preliminary safety ecaluation showed that the toxic of this granule was low.(4)The basic diet added 2.0%of Sijunzi flavored granules feeding 30d can better promote weaning Rex rabbits' growth performance and keep them health. |
