Based on the polymorphism of IGF-Ⅰ, the current study was designed to investigate the effect of IGF-Ⅰgene on growth traits of yellow chicken,and tried to provide evidence for the basic genetic research on yellow chicken.Single nucleotide polymorphism of IGF-Ⅰgene was detected in 228 individuals from 4 different yellow chicken breed including Jinghai yellow chicken, Jiangcun yellow chicken, Shiqiza chicken, Liyang yellow chicken by PCR-SSCP, and two individuals were selected to directly sequence for each primer. The results showed that three genotypes, named AA, AB and BB were found to exit in each of the chicken population. As for result of sequence analysis of primer 4, a mutation (T→A) site at 264 base of exon 1 was detected, and all the yellow chicken population were in Hardy-Weinberg equilibrium state. And as for the result of sequence analysis of primer 7, a mutation (G→A) site at 528 base of exon 3 was detected, and the chi-square text (χ~2) showed that Jiangcun yellow chicken, Shiqiza chicken and Liyang yellow chicken were all in Hardy-Weinberg equilibrium state, but Jinghai yellow chicken had the opposite result. The text result showed that Hardy-Weinberg equilibrium had variety characteristic.The statistical result of primer 4 between different genotypes and chicken growth traits of different yellow chicken populations showed that: AA genotype had significant enhance effect (P﹤0.05) on the number of egg laid of 300 days in Jinghai yellow chicken population.
