| Euscaphis japonica(Thunb.)Kanitz is a species in the family Staphyleaceae which is a wild ornamental plant with beautiful flower and fruit,and is also used to be a medicinal plant.E.japonict is hard to be a large number of application because the difficulty of seed germination,the germination rate is low,the budding time is inconsistency,and cuttage survival rate is low so on.In order to solve the problem of E.japonica breeding,the plant tissue culture.technology has been applied.However,the present research mainly forcus on the common E.japonics more than varieties or cultivars.In addition to E.japonica tissue culture light environment research has not been reported.The space mutation of E.japonica tissue culture rapid propagation system was established by disinfection,initial culture,subculture,rooting culture and transplanting Study on the influence of LED light quality on space mutation of E.japonica multiplication culture and rooting culture.This research used stem segment with bud of E.japonica by variegated-leaf to study disinfection,primary explants,subculture.Set up the foundation of E.japonica Cultivars and Varieties of factory seedling.The main results were as follows.1.The rapid multiplication system of E.japonica was establish by space mutation breeding.Explant is stem segment with bud of E.japonica by space mutation breeding.The optimum disinfect condition was in the room for 5d after 75%alcohol disinfection 30s and then in 0.1%HgCl2 disinfection 8min.The suitable culture medium for primary culture was MS+1.0mg·L-1 6-BA +0.1mg·L-1 NAA which the rate of effective germination was 94.44%.The suitable culture medium for subculture was MS+1.0mg·L-1 6-BA+ 0.1mg·L-1 NAA with the proliferation value of 4.12.The suitable culture medium for rooting was 1/3WPM+0.3 mg·L-1 IBA+0.3 mg·L-1 NAA,the rate of rooting was 69.23%.The suitable transplanting substrate was peat + perlite+ sand=8:1:1,and the survival rate of transplanting was 96.67%.2.The tissue culture of E.japonica"Silver" by variegated-leaf was studied.Statistical analysis showed that 75%alcohol disinfection 30s and then in 0.1%HgCl2 disinfection 8min was suitable.The suitable culture medium for primary culture was MS+1.0mg·L-1 6-BA+0.1mg·L-1 NAA and the rate of effective germination was 92.56%.The suitable culture medium on subculture was MS+2.0mg·L-1 6-BA+0.5mg·L-1 NAA with the proliferation value of 4.07.3.The physiological characteristics of E.japonica at different LED light quality on the Space Mutation Tissue Culture Seedlings have been.investigted.Proliferation value of the blue light treatment was the highest which was 4.29.Chlorophyll a content,total chlorophyll content,carotenoid content,soluble protein content and SOD activity were the greatest in plantlets cultured under light quality of red plus blue=3:1,numerical value were 2.240mg·g-1,2.961mg·g-1,0.437mg·g-1,21.410mg·g-1 and 287.699 U·g-1 respectively.The highest Chlorophyll b content of light quality of red plus blue=5:1 treatment were 0.773mg·g-1.Soluble sugar and POD activity were the greatest in plantlets cultured under light quality.of red plus blue=7:1,42.298mg·g-1 and 3625U·g-1·min-1 respectively.The CAT activity of red light treatment reached to 25U·g-1·min-1.4.The physiological characteristics of Ejaponica by Space Mutation Tissue Culture Seedlings in rooting.under different LED light quality showed that ight quality of red plus blue=5:1 was the best condition though measuring fresh weight and dry weight of plantlets cultured,numerical value were 217.67mg and 46.67mg respectively.The plant height,chlorophyll a,chlorophyll b,total chlorophyll,carotenoid content and POD activity were highest in plantlets cultured under light quality of red plus blue=7:1,numerical value were 3.62cm,2.693mg·g-1,1.113 mg·g-1,3.806mg·g-1,0.503mg·g-1 and 8230U·g-1·min-1 The soluble sugar content of the white light treatment were highest reached to 46.244mg·g-1.Soluble protein content and CAT activity were highest in plantlets cultured under red light,numerical value were 18.651mg·g-1 and 13.542U·g-1 ·min-1.The light quality of red plus blue=3:1 was the best condition based on SOD activity,it was 456.35 1U·g-1.Considering the varives physiological characteristics,light quality of red plus blue=7:1 was optimum for E.japonica by Space Mutation Tissue Culture Seedlings in rooting. |
