Tree peony belonged to the Paeonia genus Paeoniaeeae,which is a deciduous shrub and unique precious germplasm resource in China.The study found that peony is a kind of oil crop with higher nutritional and health-care value in recent years.It has been found more suitable for human nutrition so far,so the development of oil peony is growing.At present,there are few reports about the cultivation and breeding of oil peony,and the research on molecular-marker assisted selection breeding(MAS)of oil peony yield has not been reported.This study based on the methods of SSR molecular markers,and analyzed the genetic diversity of the 36 peony cultivars with good oil production and ornamental quality.Through the analysis of the genetic structure of the population to determine the value of Q,the correlation between phenotypic traits and molecular markers of peony was analyzed by GLM analysis in TASSEL software to locate some related traits and find the molecular markers closely linked with the yield traits of the oil peony,providing the theoretical basis for the gene location,genetic improvement and molecular marker assisted breeding of the peony related traits.1.In this study,16 pairs of SSR primers were used to amplify by PCR,results show that 65 loci are produced during these36 peony.Loci of every marker ranged from 2 to 8,with an average of 4.06.The mean polymorphism information content value was with a range from 0.19 to 0.73,with an average of 0.50.The Shannon diversity index ranged from 0.42 to 1.59,with an average of 1.01.The observed heterozygosity ranged from 0.03 to 0.83,with an average of 0.37.The exopected heterozygosity ranged from 0.21 to 0.78,with an average of 0.56.The mean observed heterozygosity(0.37)tended to be lower than the mean expected heterozygosity(0.56).The results showed that SSR markers had high genetic diversity and the population had high genetic diversity.2.Cluster analysis was performed on 36 Peony cultivars by UPGMA method,the similarity coefficient of 0.64 materials could be divided into two groups.The first group included 35 germplasm resources,including MCJH as a group,indicating that the varieties are distantly related with the other 35 groups.At the similarity coefficient of 0.67 35 the 35 groups could be divided into two groups.The clustering results are basically consistent with the basic taxonomy.It was found that the yield characters and color of the oil were also related with clustering results.3.Association analysis between 16 SSR loci and 25 ornamental traits,including 9 yield traits,4 fruit traits and 13 growth traits of 36 Peony cultivars was performed.The results showed that 11 markers were significantly correlated with the 16 traits.Among the growth traits,9 loci were significantly correlated with 9 personality traits.Among the growth traits,9 loci were significantly correlated with 9 personality traits.Among the yield traits,6 loci were significantly correlated with 6 characters.In the fruit characters,only one character of follicles was associated with 1 locus.In the correlation analysis of the yield traits,there were 6 significant levels(P <.0.01),and the variation of SSR12 on aggregated follicles to explain the highest rate of 66.15 %.In this study,the correlation analysis between yield traits and SSR molecular markers was first applied to the oil peony.The results from this research will lay the foundation for the development of gene location,gene cloning and molecular marker assisted breeding of peony related traits,and provide a scientific evidence for the efficient exploration of other excellent genetic resources and germplasm innovation of oil peony.
