In recent years,with the development of the aquaculture industry,various feed additives and antibiotic products have been abused,thus it draws more and more concerns to the issues of food safety and the ensuing environmental safety,and it spreads the concept of new green feed additives to the public gradually.This laboratory has developed a compound micro-ecological preparation product named Gongda II which is mainly used in the cultivation of sea cucumber.However,this product is mainly composed of fermented broth.It will affect the effect of this product for its single form,inconvenience of its transportation and the short shelf life.Therefore,this thesis carried out the research on the embedding and spray drying of the probiotic Gongda II in view of the existing problems.The first part of this thesis studied the embedding process of Gongda II by choosing the embedded carrier,determining the ratio of the best embedding material,making the embedded microspheres and then observing the internal structure of the microspheres through SEM?scanning electron microscope?.Finally,the microspheres embedded in Gongda II were put into artificial wastewater for the water purification experiment.The study showed these results.?1?The modified zeolite powder was chosen as the carrier,and the best ratio of the embedded material consisted of the following parts:6%of vinyl alcohol?PVA?,3%of sodium alginate?SA?,3%of CaCl2,and 2%of zeolite powder.Besides,the copolymer microspheres were uniform in size;the water content of the microspheres was 89.84%;the bacterial cell embedding rate was 89.95%;the zeolite powder embedding rate was 93.43%.?2?It was observed through the SEM?Scanning Electron Microscopy?that there was loose spatial structure of the microspheres in which it was filamentous and connected into a network,which increased the internal contact space and facilitated material exchange.?3?This study Embedded Gongda II into microspheres and put it into artificial wastewater to determine its effect to purify water.Consequently,Gongda II had the prominent ability to remove ammonia nitrogen,chemical oxygen demand and sulfides from the artificial wastewater.Moreover,the duration of water purification was 2-4 days longer than that before embedding,which showed that the embedding treatment can improve its water purification effect.Furthermore,it studied the spray drying process of Gongda II to determine the two parameters of spray drying feed flow rate?percentage of peristaltic pump rotation speed?and inlet air temperature,and then it aimed to improve the drying efficiency by adding an appropriate amount of drying aid.It came out these results.?1?When the feed flow rate was60%and the air inlet air temperature was 210°C,the drying effect was the best.The water content of bacteria powder was 3.85%;the yield of bacterial powder was 49.15 g/L;the number of live bacteria was 4.9×108 CFU/g.?2?The ratio of?-cyclodextrin to cornstarch was1:3,and they were mixed to be used as a mixed drying aid.The best drying effect was achieved with 10%addition,and the yield and number of live bacteria were significantly increased comparing with the non-addition.In summary,after application of the embedding and spray drying process to the processing of the probiotic Gongda II,the quality of the product is significantly improved,and the comparative broth product is easy to carry,easy to preserve,and saves space,solving the problem of inconvenient transport of the broth,and preservation.Short-term problems can be promoted in the aquaculture industry.
