It is found that polysaccharides have significant antioxidant activity and relatively less toxic and side effects on the body.The research on bioactivity of polysaccharides has become a hot research topic in medical field.The purpose of this study was to study the protective effects of Codonopsis Polysaccharide?CP?and Sulfated Codonopsis Polysaccharide?SCP?on acute oxidative damage in macrophages and mice.This experiment is divided into three parts.The first part is to extract and purify CP by water decocting,alcohol precipitation and Sevage method,and sulfated and modified by chlorsulfuric acid pyridine method.The results showed that the sugar content of CP and SCP was 0.8120 mg/mL and 0.6630 mg/mL respectively,and the sulfate content of SCP was 0.3448 mg/mL.After NIR and SEM observation,sulfated modification was good.The second part is using H2O2 to stimulate RAW 264.7 cell damage caused by detection of SOD,MDA,GSH-Px,CAT,ROS,phagocytosis,cell apoptosis,NO,iNOS,IL-2/4/10,IFN-?and gamma growth state index in VC group as control,to observe the protective effects of CP and SCP of different concentrations on oxidative damage in RAW264.7 cells the.The results showed that:1.CP and SCP had an effective antioxidant effect on macrophages stimulated by H2O2;2.CP and SCP both effectively improved the ability of cell phagocytosis,of which the concentration group in SCP and the high concentration group of CP were the strongest;3.CP low concentration group and SCP concentration group effectively protect macrophages under H2O2 stimulation to reduce apoptosis;4.CP and SCP effectively promote the secretion of NO in macrophages and inhibit the secretion of iNOS;5.CP and SCP have a significant effect on the secretion of IL-2 and IFN-gamma in macrophages,and obviously inhibit the secretion of IL-4;6.CP and SCP effectively protect the growth of cells under a certain concentration of H2O2.The third part is using ethanol gavage to mice caused by acute oxidative damage,the body weight,organ index,blood routine,blood biochemistry,SOD,MDA,GSH-Px,CAT,ROS,IL-2/4/10,IFN-?,RT-PCR detection,histological observation method,with VC as control group,to observe the protective effect of CP and SCP of different concentrations on acute oxidative injury in mice.The results of the experiment were as follows:1.the blood routine index showed acute inflammation and acute oxidative damage in mice fed with ethanol;2.CP and SCP have anti-inflammatory effects on acute oxidative injury in mice;3.the biochemical index of blood showed that the mice had acute oxidative damage,and CP and SCP had protective effect on mice with acute oxidative damage;4.CP and SCP have obvious effect on the secretion of IL-2 and IFN-?in acute oxidative damaged mice,and have obvious inhibitory effect on the secretion of IL-4;5.CP and SCP may be involved in the regulation of Keap1-Nrf2-ARE pathway in the antioxidant system in mice;6.The liver and kidney tissue sections of mice were observed under HE staining microscope.The liver tissue of the model group had obvious degeneration,and there was no degeneration in the blank group and the other groups.
