| As the most important micro ecological environment in the body,the intestinal tract is an important place for digestion and absorption of nutrients,which has been paid more and more attention.The dynamic balance of gut microbiota indicates the state of health.The objective of this paper is mainly to research the effect of heat stress on the diversity and richness of gut microbiota,abundance of microbial community,heat shock protein,cortisol,blood biochemical index,fatty acid of egg,and the regulating action of Chinese Medicine in laying hens.It was to further explore the mechanism o f Chinese Medicine on the thermal stress model.This can provide ascientific basis for livestock and poultry production of Chinese medicine.The health,no significant difference of Roman layers at the age of 35 weeks were randomly divided into 3 groups,Traditional C hinese Medicine group,high temperature group,blank control group,and 10 in each group.The temperature of Traditional C hinese Medicine group and high temperature group were 35±2℃,and the blank control group was 24±2℃.Traditional C hinese Medicine group was added with the Traditional Chinese Medicine in the feed.O n 1d,7d,14 d,28d,the blood sampling through subclavian vein were collected.The blood sampling was to detect heat shock protein,cortisol,blood biochemical indexes.On 1d,3d,7d,14 d,28d,the feces were collected.The feces was to sequence the gut microbiota by Illumina Miseq,and it was to analyze the diversity and richness of gut microbiota and abundance of microbial community.On 14 d and 28 d,6 eggs were randomlyselected to detect the content of fatty acid.The results were as follows:On days 1,3,14 and 28,the chao1,observed-species,PD-whole-tree,Shannon index of high temperature group were reduced compared with Traditional Chinese Medicine group and the blank control group,and O n days 1,14 showed significant difference in the diversity and richness of gut microbiota by heat stress(P<0.05).PCA and PCo A analysis showed no obvious difference betweenhigh temperature group and blank control group in the gut microbiota in the initial of study.But with the increase of heat stress time,the heat stress significantly affectedthe structural of gut microbiota,and it was significantly improvedwith the treatment of Traditional Chinese Medicine.The relative abundance of Bacteroidaceae,Pasteurellaceae,Lachnospiraceae were significantly increased and Enterococcaceae,Lactobacillaceae were decreased by heat stress.These changes were inhibited by Traditional Chinese Medicine and heat stress was alleviated.The relative abundance of Bacteroides,Oscillospira,Ruminococcus were significantly increased and Enterococcus,Lactobacillus were decreased by heat stress.Althoughthe relative abundance of Veillonella showed no difference among the three groups,Traditional Chinese Medicine inhibited the changes by heat stress on Veillonella.From days 7,the serum phosphorus in layers were significantly reduced by heat stress(P<0.05).At days 28,Traditional Chinese Medicine significantly inhibited the decrease of serum phosphorus by heat stress(P<0.05).Duringthe whole experiment,the content of serum calciumin layers were significantly reduced by heat stress(P<0.05).At days 7,the content of serum calcium in Traditional C hinese Medicine group was significantly higher than that in the high temperature group(P<0.05).At days 1 and 7,the TP content of Traditional C hinese Medicine group and the blank control group were significantly increased compared with high temperature(P<0.05).At days 1,7,14,the ALB content of Traditional C hinese Medicine group and the blank control group were significantly increased compared with high temperature(P<0.05).At days 28,the ALB in serum were significantly reduced by high temperature(P<0.05).At days 1,7,compared with Traditional Chinese Medicine group and blank control group,the content of GLU were significantly increased in high temperature group(P<0.05).And compared with blank control group,the content of GLU were significantly decreased in high temperature group(P<0.05).At day 1,the content of TC were significantlyreduced by high temperature(P<0.05).At days 1,7 and 28,the contents of HSP70 and cortisol in the high temperature group were significantly improved with the Traditional C hinese Medicine group and the blank control group(P<0.05).At days 7,the content of serum cortisol were significantly improved byheat stress(P<0.05).At days 14,the content of stearic acid in egg were significantly improved by high temperature(P<0.05).The content of oleic acid and linoleic acid in egg were significantly improved by Traditional Chinese Medicine(P<0.05).At days 28,the content of palmitic acid in egg with Traditional C hinese Medicine were significantly improved compared with the blank control group(P<0.05). |
