Blueberries are clustered shrubs.There are two ways of propagation: vitro and cutting productive.Their reproductive modes and technical requirements are special.In this study,the seedlings and cutting seedlings of the representative “Legacy”and“Bright-well” two blueberry varieties are select from four aspects of leaf and root anatomy,growth and development,physical and chemical characteristics and resistance.Effects of growth and physiological differences of blueberry seedlings way,provide a theoretical basis for the industrial production of high quality seedling of blueberry.The results are as follows:(1)During the growth of "Bright-well" and "Legacy" cuttings and vitro seedlings,the height of the seedlings is two peak periods,from mid May to mid June and mid July to mid August,and the height of ground diameter is from mid May to the middle of June.The root dry weight,stem weight,dry weight and total dry weight of the cutting and vitro seedlings are significantly different(P < 0.05).The dry weight and total dry weight of the cutting seedlings and the vitro seedlings are significantly different(P < 0.05),and the biomass of the tissue culture seedlings is larger than the cutting seedlings;the growth of the blueberry seedlings is analyze by principal component analysis.The results show that the growth of the "Bright-well" vitro seedlings is the best,followed by the "Bright-well" cutting seedlings.The growth status of the "Legacy" vitro seedling is third,and the growth of the "Legacy" cutting seedlings is in the worth worst condition.(2)The diurnal variation of net photosynthetic rate is "double peaks curve".In the different breeding ways,the net photosynthetic rate,chlorophyll a,chlorophyll b content,nitrate nitrogen content and root activity of the 4 seedlings are significantly different(P<0.05),and the difference of stoma conductance is not significant(P>0.05).The comprehensive evaluation of physiological and biochemical characteristics of blueberry seedlings under different reproductive formula by principal component analysis.As a whole,the vitro seedlings are stronger than the cutting seedlings,and the "Legacy" is stronger than the "Bright-well".(3)Under the drought stress,the relative water content and chlorophyll content o f blueberry seedlings under different breeding methods decrease with the increase of t he number of stress days.The content of malondialdehyde(MDA),proline content(Pr o)and relative conductivity of leaves increase with the increase of the number of days,but the increase in different days is not one.The comprehensive evaluation method of the average value of membership function is used to evaluate the drought resistance.The ranks of the drought resistance are as follows: "Bright-well" vitro,“Legacy”vitr o,“Legacy”cutting seedling,"Bright-well" cutting seedlings.It can be seen that thedrought resistance vitro seedlings are stronger than the cutting seedlings.(4)Under the low temperature stress,relative conductivity increases with the decrease of stress temperature,and the relative conductivity increases gradually,showing a "S" type change.The function expression between temperature and relative conductivity is obtained by Logistic equation.The semi lethal temperature of 4 kinds of seedlings is obtained by expression,and the semi lethal temperature is between-4.48?and-7.02?.From strong to weak,the ranks of the drought resistance are as follows: "brilliant" tissue culture seedlings,"Legacy" cutting seedlings and "Bright-well" cutting seedlings,and the resistance of 4 seedlings is evaluated by the membership function combined with the drought resistance.The results show that the resistance of the vitro seedlings is stronger than the cutting seedlings.(5)The paraffin section and paraffin oil film method are used to make the paraffin section and the paraffin oil film method is used to observe the stoma in different months.The results show that there are significant differences in the upper epidermis,the lower epidermis,the palisade tissue,the sponge tissue and the leaf thickness of the 4 seedlings in different months(P < 0.05)the structure of the leaves in the same month is more than the "Bright-well" seedlings,and the number of cuttings in the thickness of each tissue is larger than that of the tissue culture.According to the comprehensive evaluation of principal component analysis,the ranks of the anatomical characteristics of the leaves are as follows: "Bright-well" cutting seedlings,"Bright-well" vitro,“Legacy” cutting seedling,“Legacy”vitro.The results show that the tissue culture of the leaf anatomical structure is stronger than the cuttage.
