In this study,blueberry seedlings,that is in tissue culture stage and cutting propagation stage,were used to analyze the effects of different LED light quality on the growth and development,and the formation of phenolic compounds of leaf.The effects of red plus blue color of LED light on the proliferating culture and rooting culture were studied by using Vaccinium ulignosum Linn.;With blueberry varieties "legacy" as the test materials,we analyzed the effect of mixeing color supplement LED light and using oligosaccharides on growth and physiological state of blueberries.Aiming to find out the light conditions for the rapid propagation of the tissue seedlings and the supplementary light conditions suitable for the blueberry cuttings.The main findings are as follows:1.Compared with the traditional white fluorescent lamp(CK),the same illumination of red plus blue LED light source can significantly improve the biomass and multiplication coefficient of Vaccinium ulignosum Linn..According the comprehensive effect,1B1R light source(50%Red plus 50%Blue)is the best,which biomass accumulation increased by 363.02%.In the 3B1R treatment(25%Red plus 75%Blue),the content of chlorophyll a in proliferating seedlings(0.14±0.02mg/g·FW)was significantly lower than that of other treatments,discreased by 54.79%,but the sucrose(9.22±0.37mg/g·FW),fructose(20.04±0.78mg/g·FW)and glucose contents(16.63±0.69mg/g·FW)were significantly higher than other treatments.2.There were significant differences in plant height(50.93±5.67mm),stem diameter(0.76±0.047mm),leaf area(0.37±0.02cm2)and root growth of other lights compared with the 3B1R light quality.According the comprehensive effect,3B1R light source is the best.The chlorophyll content of rooting seedlings that were under LED light is significantly higher than that of CK,with the increase of the proportion of blue light,the chlorophyll content of the rooting seedlings in vitro was gradually increased,and reached the maximum under 3B1R treatment(2.82±0.27mg/g-FW).The content of sucrose(13.92±1.30mg/g FW)and glucose(10.77±0.49mg/g FW)in 1B1R light source was higher than that in other treatment.3.The main components of anthocyanins in the Vaccinium ulignosum Linn in vitro:Petunidin-3-O-galactoside,Petunidin-3-O-glucoside,Delphinidin-3-O-arabinoside.In proliferating medium,the content of anthocyanin in the proliferating Vaccinium ulignosum Linn in vitro also increased with the increase of the proportion of blue light in the mixing light,the content of Petunidin-3-O-glucoside and Delphinidin-3-O-arabinoglycans increased gradually with the increase of the proportion of blue light,which was significantly higher than that of other treatments under 3B1R treatment.In rooting medium,the anthocyanin content of rooting seedlings under red and blue mixed color LED light source was significantly higher than that of CK,which was mainly affected by the content of Petunidin-3-O-glucoside and Delphinidin-3-O-arabinoglycans.4.In this study,it was found that oligosaccharides had no significant effect on the morphological and physiological characteristics of blueberries that is legacy.BR and BRG supplement light source can significantly promote the growth of legacy,and can promote the synthesis of phenolic compounds in blueberry leaves,but the difference between the two light source treatment is not significant.5.Red plus blue light is the key to the content of anthocyanins in blueberry leaves.The light intensity and the ratio of red and blue light can significantly affect the synthesis of anthocyanins in leaves.The anthocyanin fractions of blueberry leaves were basically the same with Vaccinium ulignosum Linn in vitro,and only one of anthocyanin fractions were compared with wild blueberry tissue culture seedlings:Delphinidin-3-O-galactoside The synthesis of Delphinidin-3-O-galactoside is mainly affected by the light intensity,and the ratio of red and blue light does not significantly affect its synthesis.
